Gut dysbiosis and bile acid (BA) metabolism disturbance are involved in the pathogenesis of ulcerative colitis. This study aimed to investigate effect fucoidan on BA gut microbiota dextran sulfate sodium-induced colitis mice. Our results showed that effectively suppressed colonic inflammation repaired barrier. In addition, increased relative abundance Lachnospiraceae family, such as Turicibacter, Muribaculum, Parasutterella, Colidextribacter, followed by an increase short-chain fatty acids, especially butyrate. Moreover, modulated elevating cholic acid, ursodeoxycholic deoxycholic lithocholic decreasing β-muricholic which led activation FXR TGR5 further enhanced barrier inflammation. revealed alleviating was largely mediated microbiota, confirmed fecal transplantation experiment. Abstract Inflammatory Bowel Disease (IBD), a chronic nonspecific intestinal inflammatory disease, is comprised of Ulcerative Colitis (UC) and Crohn’s (CD). IBD closely related to systemic reaction affects the progression many extraintestinal diseases. As one representative bacteria for probiotic-assisted therapy in IBD, multiple strains Lactobacillus have been proven alleviate damage strengthen immunological barrier, epithelial cell mucus barrier. also spares no effort alleviation IBD-related diseases such as Colitis-associated Colorectal cancer (CAC), Alzheimer’s (AD), Depression, Anxiety, Autoimmune Hepatitis (AIH), so on via gut-brain axis gut-liver axis. Disorder in bile acid (BA) metabolism is known to be an important factor contributing diarrhea. However, the pathogenesis of BA disorder-induced diarrhea remains unclear. The colonic pool and microbiota between health piglets diarrheal were compared. Fecal transplantation various cell experiments further indicated that chenodeoxycholic (CDCA) metabolic disorder produced CDCA-3β-glucuronide, which main cause Non-targeted metabolomics uncovered inhibition glucuronidation by Lactobacillus reuteri (L. reuteri) through deriving indole-3-carbinol (I3C). In vitro, gene involved reduction induced-diarrhea screened RNA transcriptomics sequencing, activation pathway FXR-SIRT1-LKB1 alleviate P53-mediated apoptosis proposed vitro multifarious siRNA interference, CO-IP, immunofluorescence, so on, mechanism was also verified a variety mouse models. Here, we reveal for first time core derived I3C represses gut epithelium glucuronidation, particularly 3β-glucuronic CDCA production, reaction mediated host UDP glucuronosyltransferase family 1 member A4 (UGT1A4) necessary induced Mechanistically, L. activates aryl hydrocarbon receptor decrease UGT1A4 transcription CDCA-3β-glucuronide content, thereby upregulating signal. Antioxidant polyphenols from plants are potential dietary supplementation to alleviate early weaning‐induced intestinal disorders in piglets. Recent evidences showed polyphenol quercetin could reshape gut microbiota when it functioned as anti‐inflammation or antioxidation agents rodent models. However, the effect of on and weanling piglets, along with role this effect, both remain unclear. Here, we determined quercetin’s attenuating diarrhea, damage, redox imbalance, well by transferring quercetin‐treated fecal recipient The results that decreased piglets’ scores improved damage increasing tight junction protein occludin, villus height, height/crypt depth ratio but crypt epithelial apoptosis (TUNEL staining). Quercetin also increased antioxidant capacity indices, including total capacity, catalase, glutathione/oxidized glutathione disulfide oxidative metabolite malondialdehyde jejunum tissue. Two weeks later, (250 mg/kg) or carboxymethylcellulose (CMC) administrated once daily by gavage twelve weeks. We found that alleviated fasting blood glucose (FBG) triglyceride (TG) as well glycosylated hemoglobin (HbA1c) levels; increased insulin (FINS); improved cardiac dysfunction; ameliorated myocardial hypertrophy, fibrosis injury; suppressed oxidative stress, inflammasome necroptosis; but SIRT3 expression STZ-induced mice. Neonatal rat cardiomyocytes pre-treated (80 μM) without high (HG) stimulation. results showed attenuated cell damage inhibited necroptosis Moreover, above protective DCM unavailable SIRT3-KO mice, implying promising medical potential treatment.
