Abstract Osteoarthritis (OA) is a common degenerative joint disease characterized by progressive cartilage degradation and inflammation. In recent years, mesenchymal stem cells (MSCs) derived exosomes (MSCs-Exo) have attracted widespread attention for their potential role in modulating OA pathology. However, the unpredictable therapeutic effects of been significant barrier to extensive clinical application. this study, we investigated whether fucoidan-pretreated MSC-derived (F-MSCs-Exo) could better protect chondrocytes osteoarthritic joints elucidate its underlying mechanisms. order evaluate F-MSCs-Exo osteoarthritis, both vitro vivo studies were conducted. MiRNA sequencing was employed analyze MSCs-Exo F-MSCs-Exo, enabling identification differentially expressed genes exploration mechanisms behind protective osteoarthritis. Compared MSCs-Exo, demonstrated superior effectiveness inhibiting inflammatory responses extracellular matrix rat chondrocytes. Moreover, exhibited enhanced activation autophagy revealed that miR-146b-5p emerged as promising candidate mediator chondroprotective function with TRAF6 identified downstream target. conclusion, our research results demonstrate encapsulated effectively inhibits activation, thereby suppressing degradation, while promoting chondrocyte protection cells. The increasing incidence of colitis and the side effects its therapeutic drugs have led to search for compounds natural origin, including phenolics, as new treatments colitis. In this study, potential mechanism Dendrobium officinale leaf phenolics (DOP) on relief dextran sulfate sodium (DSS)-induced was explored. results showed that DOP treatment 36 days reduced symptoms caused by DSS, reduction disease activity index alleviation colonic tissue damage. addition, downregulated expression key proteins TLR4/NF-κB signaling pathway production inflammatory cytokines. Furthermore, could enhance tight junction ZO-1, Occludin, Claudin-1 restore intestinal mucosal barrier function. also effectively regulates disordered flora enhances short-chain fatty acids, which is beneficial in modulating gut internal environmental homeostasis, inhibiting inflammation, restoring barrier. These findings indicated can ameliorate DSS-induced chronic regulating microbiota, barrier, it a promising ingredient from D. officinale.

The prevalence of non-alcoholic fatty liver disease (NAFLD) is increasing annually, and emerging evidence suggests that the gut microbiota plays a causative role in development NAFLD. However, NAFLD remains unclear warrants further investigation. Thus, C57BL/6J mice were fed high-fat diet (HFD), we found HFD significantly induced obesity increased accumulation intrahepatic lipids, along with alterations serum biochemical parameters. Moreover, it was observed also impaired barrier integrity. It revealed via 16S rRNA gene sequencing microbial diversity, which enriched Colidextribacter, Lachnospiraceae-NK4A136-group, Acetatifactor, Erysipelatoclostridium. Meanwhile, reduced abundance Faecalibaculum, Muribaculaceae, Coriobacteriaceae-UCG-002. predicted metabolic pathways suggest enhances chemotaxis functional activity associated flagellar assembly, while risk intestinal pathogen colonization inflammation. And phosphotransferase system, streptomycin biosynthesis, starch/sucrose metabolism exhibited decreases. These findings reveal composition predictive functions microbiome NAFLD, corroborating association between providing novel insights into its potential application research for patients.

Diabetic nephropathy (DN) is a common microvascular complication of diabetes. Fucoidan, polysaccharide containing fucose and sulfate group, ameliorates DN. However, the underlying mechanism has not been fully understood. This study aimed to explore effects fucoidan on DN in high-fat diet-induced diabetic mice. A total 90 C57BL/6J mice were randomly assigned six groups (n = 15) as follows: normal control (NC), diabetes mellitus (DM), metformin (MTF), low-dose (LFC), medium-dose (MFC), high-dose (HFC). technique based fluorescein isothiocyanate (FITC-sinistin) elimination kinetics measured percutaneously was applied determine glomerular filtration rate (GFR). After 24 weeks, sacrificed an early stage model confirmed by GFR hyperfiltration, elevated urinary creatinine, albumin, tubulointerstitial fibrosis, hypertrophy. Huaier (Trametes robiniophila Murr) fungus that been used clinically more than 1000 years, its bioactive polysaccharide components reported to possess immunomodulatory effects, antitumour potential, renoprotective effects. In this study, we aimed examine the protective effects mechanisms (HP) against UC. Based H2O2-induced oxidative stress model HT-29 cells dextran sulphate sodium salt (DSS)-induced model, demonstrated polysaccharides significantly alleviated DSS-induced (weight loss, elevated disease activity index (DAI) scores, colonic shortening). addition, HP inhibited inflammation intestinal barrier damage.
