Abstract Ulcerative colitis (UC) is a chronic, relapsing inflammatory disorder of the gastrointestinal tract. Programmed cell death (PCD), such as PANoptosis and autophagy, involved in various inflammation- immune-related diseases. This study aimed to examine molecular signature associated immune infiltration PANoptosis- autophagy-related differentially expressed genes (DEGs) UC. Ten PANoptosis-related hub DEGs, including PDGFRB, TIMP1, MMP2, CD44, TIMP2, TGFB2, IL6, TIMP3, IL1B, HGF, four CCL2, TGFB1, PPARG, CXCR4, were identified. These with chemotaxis, wound healing positive regulation MAPK cascade. Immune analysis revealed UC patients exhibited higher immunocytes these closely related infiltrations. Through machine learning algorithms, five key candidate genes, showed good diagnostic performance distinguishing from healthy individuals. Furthermore, highly fibroblasts (IAFs) by single-cell sequencing analysis. The expression levels significant differences colon mucosa patients. Our findings indicated that autophagy or crosstalk between them might be implicated dysregulation via regulating specific cells IAFs interacting signals chemotaxis signaling pathways.

Purpose: Polygala tenuifolia Willd. (PT), a traditional Chinese medicinal plant extensively employed in managing Alzheimer's disease, exhibits notable gastrointestinal side effects as highlighted by prior investigations. In contrast, Magnolia officinalis Rehd. et Wils (MO), remedy for ailments, shows promising potential ameliorating this adverse effect of PT. The objective study is to examine the underlying mechanism MO alleviating Methods: Hematoxylin-eosin (H&E) staining was used observe structural damage zebrafish intestine, and enzyme-linked immunosorbent assay (ELISA) detect levels inflammatory factors oxidative stress. integrity intestinal tight junctions examined using transmission electron microscope (TEM). Moreover, expression barrier genes PI3K/AKT/NF-κB signaling pathway-related determined through quantitative real-time PCR. changes microbial composition were analyzed 16S rRNA metagenomic techniques. Results: effectively ameliorated pathological gene expression, significantly alleviated injury reducing cytokines IL-1β, IL-6, TNF-α, inhibiting activation pathway. Furthermore, could increase relative abundance beneficial microorganisms ( Lactobacillus, Blautia Saccharomyces cerevisiae ), reduce pathogenic bacteria Plesiomonas Aeromonas ). Conclusion: PT-induced injury, its may be related inhibition pathway regulation flora. Keywords: polygala tenuifolia, PI3K/AKT/NF-κB, gut microbiota, inflammation,

Spinal cord injury (SCI) remains a formidable challenge, lacking effective treatments. Following SCI, neural stem cells (NSCs) migrate to SCI sites, offering potential avenue for nerve regeneration, but the effectiveness of this intrinsic repair mechanism suboptimal. Salidroside has demonstrated pro-repair attributes in various pathological conditions, including arthritis and cerebral ischemia, ability curtail early-stage inflammation following SCI. However, specific role salidroside late-stage processes less defined. In investigation, we observed that continuous treatment mice improved motor function recovery. Immunofluorescence-staining corroborated salidroside's capacity stimulate regeneration remyelination, suppress glial scar hyperplasia, reduce activation neurotoxic A1 astrocytes, facilitate NSCs migration towards injured region. Mechanistically, vitro experiments elucidated significant restraining astrocyte proliferation polarization. It was further established astrocytes hinder while inducing their differentiation into astrocytes. effectively ameliorated inhibition through diminishing c-Jun N-terminal kinase (JNK) pathway phosphorylation restored neurons by suppressing signal transducer activator transcription 3 (STAT3) pathway. summary, our findings suggest holds promise as therapeutic agent traumatic treatment.

Ulcerative colitis (UC) is a chronic, nonspecific inflammatory disorder characterized by symptoms such as abdominal pain, diarrhea, hematochezia, and urgency during defecation. While the primary site of involvement colon, UC can extend to encompass entire rectum colon. The causes development mechanisms are still not well understood; nonetheless, it currently held that factors including environmental influences, genetic predispositions, intestinal mucosal integrity, gut microbiota composition, immune dysregulation contribute its development. Dysregulated responses pivotal in pathophysiology UC, these aberrant considered key contributors disease onset. In patients with cells become hyperactive erroneously target normal tissue, resulting cascades damage mucosa. therapeutic strategies employed for include immunosuppressive agents aminosalicylates corticosteroids. However, treatments often prove costly carry significant adverse effects — imposing considerable burden on patients. Traditional Chinese Medicine (TCM) has attracted worldwide attention because multi-target approach, minimal side effects, cost-effectiveness, favorable efficacy profiles. this review, ways which TCM modulates treatment ulcerative have been outlined. Research into modalities modulating pathways yielded promising advancements, individual herbs, herbal formulations, their derivatives, summarized. utilized treat system plays role regulating homeostasis. It imperative facilitate large-scale evidence-based medical research promote clinical application management UC.
