The efficacy of deferiprone (DFP) in reducing liver iron content is still inadequate. Ethanolic extract Phaleria macrocarpa L. (PM) fruit showed chelation activity and hepatoprotective effects. This study aimed to evaluate the combination PM reduce liver. Rats were divided into 6 groups: normal, overload (IO), IO+DFP, IO+PM extract, IO+PM+DFP or half dose. induced with iron-dextran injections over 8 weeks, therapy starting from week 4. Measurements included hepatic iron, malondialdehyde (MDA), GSH, TNF-α levels, GPx4 mRNA expression, plasma ALT AST activity, histopathology. While none treatments significantly reduced treatment PM+DFP dose greatest reduction improved MDA, ALT, AST, levels compared DFP monotherapy. GSH not altered by therapy.

Iron, an essential mineral in the body, is involved numerous physiological processes, making maintenance of iron homeostasis crucial for overall health. Both overload and deficiency can cause various disorders human diseases. Ferroptosis, a form cell death dependent on iron, characterized by extensive peroxidation lipids. Unlike other kinds classical unprogrammed death, ferroptosis primarily linked to disruptions metabolism, lipid peroxidation, antioxidant system imbalance. Ferroptosis regulated through transcription, translation, post-translational modifications, which affect cellular sensitivity ferroptosis. Over past decade or so, diseases have been as part their etiology, including cancers, metabolic disorders, autoimmune diseases, central nervous cardiovascular musculoskeletal Ferroptosis-related proteins become attractive targets many major that are currently incurable, some regulators shown therapeutic effects clinical trials although further validation potential needed. Therefore, in-depth analysis its molecular mechanisms may offer additional strategies prevention treatment. In this review, we discuss significance contribution etiology development along with evidence supporting targeting approach. Lipid peroxidation (LP) leads to changes in the fluidity and permeability of cell membranes, affecting normal cellular function potentially triggering apoptosis or necrosis. This process is closely correlated with onset many diseases. Evidence suggests that phenolic hydroxyl groups food-borne plant polyphenols (FPPs) make them effective antioxidants capable preventing diseases triggered by membrane LP. Proper dietary intake FPPs can attenuate oxidative stress, especially damage phospholipids, activating Nrf2/GPx4 pathway. Nuclear factor E2-related 2 (Nrf2) an stress antagonist. The signaling pathway regulated Nrf2 a defense transduction organism against external stimuli such as reactive oxygen species exogenous chemicals. Glutathione peroxidase 4 (GPx4), under regulation Nrf2, only enzyme reduces lipid peroxides specificity, thus playing pivotal role regulating ferroptosis counteracting stress. study explored mechanism, antioxidant activity FPPs, mechanism It also highlighted bioprotective properties LP its associated mechanisms, including (i) activation pathway, GPx4 serving central target protein, (ii) activities, leading reduction production ROS other peroxides, (iii) effects on downstream phospholipid structure. In conclusion, play crucial natural However, further in-depth analysis coregulation multiple pathways required, combined these mechanisms need evaluation experimental models. Human trials could provide valuable insights into new directions for research application.

Ulcerative colitis is a chronic intestinal disease linked to barrier damage, ferroptosis and dysbiosis. Coix lacryma-jobi natural food with food-medicine homology, whose seed-derived oil (Coix seed oil, CSO) has been shown anti-inflammatory activity in vitro. Here, the effects mechanisms of CSO on ulcerative (UC) vivo are systematically investigated. Firstly, UC mice was replicated by 3% DSS, assessed efficacy observing fecal occult blood, colon length, DAI score pathological histomorphological changes tissues. The barrier-protective were observed AB staining qRT-PCR. Secondly, biological targets obtained from TCMSP database Swiss Target Prediction database, downloaded FerrDb platform, UC-related GEO intersection above three taken obtain "CSO-UC-Ferroptosis" targets, which analysed GO KEGG enrichment, GSEA analysis, immune cell infiltration validation. Finally, core genes molecular docking potential active components CSO. In order further verify effect ferroptosis, GPX4 agonist RSL-3 used stimulate vivo, levels Iron, MDA SOD measured, immunohistochemistry detect tight junction proteins protein mice. Besides, evaluated intercellular junctions tissues each group under electron microscope. addition, 16sRNA sequencing performed contents observe flora improved physiological parameters, reduced inflammation response regulated restored gut microbiota balance Bioinformatics results showed that G6PD, ABCC1 at CSO, also demonstrated similar expression DSS-induced models vivo. Our findings demonstrate for first time ameliorated regulating mice, suggesting as promising candidate treatment warranting investigation.

Large bone defects, often arising from trauma or infection, pose a considerable therapeutic challenge due to their limited capacity for spontaneous healing, thus requiring graft materials effective reparative procedures. The persistence of inflammation and elevated levels reactive oxygen species (ROS) within these defect sites significantly impede regeneration process. Addressing this, an injectable hydrogel system with ROS-responsive functionality is developed, specifically tailored the high ROS microenvironment characteristic defects. This incorporates hyaluronic acid functionalized dopamine introduce catechol moieties, employs 4-formylphenylboronic as crosslinking agent form dynamic matrix (HAC) carboxymethyl chitosan.
