As a mechanism of cell death, ferroptosis has gained popularity since 2012. The process is distinguished by iron toxicity and phospholipid accumulation, in contrast to autophagy, apoptosis, other death mechanisms. It implicated the advancement multiple diseases across body. Researchers currently know that osteosarcoma, osteoporosis, orthopedic disorders are caused NRF2, GPX4, star proteins. effective relief osteoarthritis symptoms from deterioration been confirmed clinical treatment with inhibitors. A chemical investigation of the soil-derived fungus Trichocladium sp. XZ8 led to isolation five new indole alkaloids, trichindoles A–E (1–5), with diverse architectures, along seven known analogues (6–12). Their structures were elucidated by extensive spectroscopic data analysis, and their absolute configurations determined single-crystal X-ray diffraction modified Mosher's method. Compound 1 is a polyketide-alkaloid hybrid incorporating rare succinimide motif, compound 2 represents first example dimeric isopentenyl indole-containing alkaloid bridged propane-1,2-diol moiety. Compounds 1, 4, 8, 11, 12 showed significant neuroprotective effects against RSL3-induced ferroptosis in PC12 cells at 10 μM. Moreover, 4 might ameliorate through regulation SLC7A11 pathway ferritinophagy, suggesting potential as promising lead compounds for treatment neurodegenerative diseases.

Hepatocellular carcinoma (HCC) has a high morbidity and mortality rate, the survival rate of HCC patients remains low. Animal medicines have been used as potential therapeutic tools throughout long history due to their different structures biologically active substances with affinity human body. Here, we focus on effects mechanism action animal-derived natural products against HCC, which were searched in databases encompassing Web Science, PubMed, Embase, Science Direct, Springer Link, EBSCO. A total 24 from 12 animals summarized. Our study found that these potent anti-hepatocellular effects. The involving apoptosis induction, autophagy anti-proliferation, anti-migration, anti-drug resistance via phosphoinositide 3-kinase (PI3K)/protein kinase B (Akt)/mammalian target rapamycin (mTOR), Ras/extracellular signal regulated kinases (ERK)/mitogen-activated protein (MAPK), Wnt/β-catenin, Janus (JAK)/signal transducer activator transcription (STAT) pathways. Huachansu injection sodium cantharidate clinical applications good efficacy. We review derivatives treatment date summarize application prospect toxic side effects, hoping provide reference for drug development HCC.

Obesity is increasingly prevalent worldwide and linked to metabolic diseases, such as insulin resistance (IR) type 2 diabetes mellitus (T2DM), due excessive free fatty acids (FFAs). Although lifestyle changes are effective, they often prove be insufficient initial treatments for obesity. Additionally, while surgical pharmacological interventions available, not entirely safe or effective. Recently, interest has grown in utilizing food waste plant-derived phenolic compounds their health benefits, presenting a promising avenue managing obesity its related disorders. Indeed, many studies have examined the potential inhibitory effects of natural extract on adipocyte differentiation lipid accumulation. This study focused evaluation standardized extracts obtained from red oranges olive leaf 3T3-L1 murine pre-adipocyte functionality. Red orange (ROE) (OLE), alone combination, were tested assess anti-obesity anti-inflammatory effects, well therapeutic benefits. Three vitro models established investigate (I) differentiation; (II) mature hypertrophic adipocytes challenged with palmitic acid (PA) erastin (ER), respectively; (III) erastin-induced cytotoxicity pre-adipocytes.
