Aurothiomalate (AuTM) is an FDA-approved antiarthritic gold drug with unique anticancer properties. To enhance its activity, we prepared a bioconjugate human apoferritin (HuHf) by attaching some AuTM moieties to surface protein residues. The reaction of excess yielded single adduct, that was characterized ESI MS and ICP-OES analysis, using three mutant ferritins trypsinization experiments. adduct contains ~3 atoms per ferritin subunit, arranged in small cluster bound Cys90 Cys102. MD simulations provided plausible structural model for the cluster. evaluated pharmacological properties found be significantly more cytotoxic than free against A2780 cancer cells mainly due higher uptake. NMR-metabolomics showed HuHf induced qualitatively similar changes treated cells, indicating effects on cell metabolism are approximately same, agreement independent biochemical In conclusion, have demonstrated here molecularly precise formed between exhibits far superior drug, while retaining key mechanistic features. Evidence can serve as excellent carrier this metallodrug.

The rising demand for nondairy and nonanimal protein sources has increased plant-based beverages (PBB) consumption. However, research on their functional properties, metabolic profile, discrimination potential is limited. This study evaluated the of proton nuclear magnetic resonance (1H NMR) spectroscopy as an authentication method to discriminate milk (cow goat) PBB macro-groups, including soy-based, fruit-based (almond coconut), cereal-based (rice oat) beverages, based profile. A total 22 (soy-, almond-, coconut-, rice-, oat-based beverages), 4 cow milk, goat cartons were analyzed with 1H NMR obtain Relevant metabolites macro-groups identified through Mann-Whitney U test partial least squares-discriminant analysis. Results revealed that uridine diphosphate, glucose, adenosine key identification milk. At same time, choline guanosine emerged important markers different macro-group detection. In addition, lactose played a significant role in differentiating from PBB. conclusion, these findings represent initial step toward applying nutritional analysis PBB, opening door further into authenticity profiling.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder and represents the most common cause of dementia in elderly population worldwide. Currently, there no cure for AD, continuous increase number susceptible individuals poses one significant emerging threats to public health. However, molecular pathways involved onset progression AD are not fully understood. This information crucial developing less invasive diagnostic instruments discovering novel potential therapeutic targets. Metabolomics studies complete ensemble endogenous exogenous metabolites present biological specimens may provide an interesting approach identify alterations multiple biochemical processes associated with evolution. In this mini review, we summarize results from metabolomic conducted using nuclear magnetic resonance (NMR) spectroscopy on human samples (blood derivatives, cerebrospinal fluid, urine, saliva, tissues) patients. We describe metabolic identified patients compared controls diagnosed mild cognitive impairment (MCI). Moreover, discuss challenges issues application NMR-based metabolomics context research.

Solution was metabolomic profiles extracts. By combining proteomic analyses tissues, found increased anaerobic glycolysis, accompanied by an altered citric acid cycle oxidative phosphorylation inflamed colons; these changes associate inflammation-induced hypoxia taking place tissues. Different colitis states also characterized significantly extracts, attributable both dysbiosis characteristic as well dysregulated Strong distinctive signatures can be detected before onset symptoms.
