ABSTRACT Background Neurodegenerative diseases pose significant health challenges in the 21st century, with increasing morbidity and mortality, particularly among elderly population. One of key factors contributing to pathogenesis these is disrupted crosstalk between mitochondria endoplasmic reticulum. Mitochondria‐associated membranes (MAMs), which are regions where ER interfaces mitochondria, serve as crucial platforms facilitating communication organelles. Objectives This review focuses on structural composition functions MAMs highlights their roles. Additionally, this review, we summarize relationship MAM dysfunction various neurodegenerative diseases, including Alzheimer's disease, Parkinson's others. The involvement proteins such Sig‐1R, IP3R, VAPB maintaining ER‐mitochondrial emphasized. Conclusion Through analyzing effects provide newest insights potential therapeutic targets for treatment debilitating conditions.
