Porcine deltacoronavirus (PDCoV) and transmissible gastroenteritis coronavirus (TGEV), the two causative agents of porcine diarrhea, have been reported to be at risk cross-species transmission, including humans. However, potential host range in which these CoVs interact remains unclear. We screened 16 animal counterparts for aminopeptidase N (APN), receptor PDCoV TGEV, found that APNs from eight 17 animals could bind receptor-binding domains (RBDs) TGEV. Furthermore, RBDs mediate cellular infection by both viruses. Dog APN (dAPN) has identified as with highest capability virus infection. further resolved complex structures dAPN bound RBD/TGEV RBD, respectively, establishing its divergent modes. R325 an important residue RBD-dAPN interaction, central role Q746 T749 interaction TGEV RBD. These findings provide molecular basis transmission shed light on future surveillance CoVs.

Canine parvovirus (CPV) and canine enteric coronavirus (CCoV) are primary viral pathogens responsible for causing diarrhea in carnivores. CCoV infection generally induces mild enteritis, whereas infections solely by CPV or coinfections involving both frequently result severe can lead to fatal outcomes affected animals. This study investigated raccoon dogs farmed China between 2018 2023. Among the 133 small intestine tissue samples from with diarrhea, 107 of (80.5%) were nucleic acid positive, CPV‐2 CPV‐2a subtypes. The amplicons maintained I418T, S927A, T27S. Moreover, subtype possessed G300S, N562V, Y573F mutations compared dog‐ori CPV‐2a. 30 (22.6%) positive CCoV, containing CCoV‐IIa CCoV‐IIb Of these, 25 (83.3%) coinfected CCoV. In alone, inflammatory infiltration was observed lamina propria intestine. Immunohistochemical staining revealed that antigens mainly detected crypts Furthermore, coronavirus‐like particles samples. Next, we analyzed detailed binding model dog‐origin (RD‐ori) host aminopeptidase N (APN). receptor‐binding domain (RBD) RD‐ori showed robust dog APN but not human APN. Our results demonstrated emerging northern China. Further investigations should be performed monitor potential evolution recombination events CCoVs.

Abstract Coronaviruses are endemic and can cause disease in a wide range of domestic animal wildlife species. The virus species Alphacoronavirus-1 comprises set diverse viruses that highly recombinogenic, including feline coronavirus type 2 (FCoV-2), which is recombinant genotype 1 (FCoV-1) canine (CCoV-2). Co-infection within host promotes viral recombination; thus, to understand the origin novel variants, it crucial identify hosts be infected with multiple alphacoronaviruses. receptor for FCoV-2 CCoV-2 aminopeptidase N (APN), APN cat (Felis catus) allowing entry FCoV-2, CCoV-2, other As wild felids genetically closely related cat, they may also susceptible these However, date, natural infection has been reported exclusively canids, not felids. In this study, we retrospectively investigated localized outbreak enteritis three captive snow leopards ( Panthera uncia) at Bronx Zoo (New York City, U.S.). Whole genome sequencing revealed shedding feces sick leopards. Phylogenetic analyses pathogenic variants circulating U.S. Europe. Comparative genetic gene from five Asian felids, leopard, high percentage identity (>95.7%). These results emphasize central role emergence alphacoronavirus.

Canine coronavirus (CCoV) is an important gastrointestinal pathogen that causes serious harm to pet dogs worldwide. Here, 1791 clinical samples were collected from in different hospitals Guangxi Province, southern China, for the 2021–2024 period and detected CCoV by a multiplex RT-qPCR. The results showed 8.43% (151/1791) of positive CCoV. Sixty-five selected amplify, sequence, analyze S, M, N genes. A sequence comparison revealed nucleotide amino acid similarities genes 94.86% 94.62%, 96.85% 97.80%, respectively. Phylogenetic analysis indicated 65 strains obtained this study belonged CCoV-II genotype, which 56 CCoV-IIa subtype 9 CCoV-IIb subtype. potential recombination event S gene sequences two strains, i.e., GXBSHM0328-34 GXYLAC0318-35, have signals.
