Fluorescent brighteners (FBs) are a group of mass-produced dyestuff chemicals that have been extensively used for decades. However, knowledge their occurrence in municipal wastewater treatment plants on large geographical scale remains unknown. Herein, we implemented the first nationwide survey wastewater-derived FBs sludge across major cities China. All 25 target were detected sludge. Ionic exhibited much higher concentrations than nonionic FBs. The total (∑25FBs) ranged from 7300 to 1,520,000 ng/g, with median 35,300 ng/g. A clear distribution significantly was found East and Central China West (p < 0.05). ∑25FBs correlated well gross domestic product (GDP) population size at provincial level 0.05), demonstrating significance anthropogenic impacts FB levels urban annual emission into is estimated be 835 tons/year, which 134 tons/year directly released sludge-applied soils. The global practice of reusing sewage sludge in agriculture and its landfill disposal reintroduces environmental contaminants, posing risks to human ecological health. This study screened from 30 Chinese cities for androgen receptor (AR) disruptors, utilizing a disruptor list the Toxicology 21st Century program (Tox21), identified 25 agonists 33 antagonists across diverse use categories. Predominantly, natural products 5α-dihydrotestosterone thymidine emerged as agonists, whereas industrial intermediate caprolactam was principal antagonist. In-house bioassays disruptors displayed good alignment with Tox21 potency data, validating employing toxicity data theoretical estimations. Potency calculations revealed two pharmaceuticals (17β-trenbolone testosterone isocaproate) most potent AR three dyes (rhodamine 6G, Victoria blue BO, gentian violet) antagonists.
