Metformin, along with its biotransformation product guanylurea, is commonly observed in municipal wastewaters and subsequent surface waters. Previous studies fish have identified metformin as a potential endocrine-active compound, but there are inconsistencies regard to effects. To further investigate the reproductive toxicity of guanylurea fish, series experiments was performed adult fathead minnows (Pimephales promelas). First, explants minnow ovary tissue were exposed 0.001-100 µM or whether compounds could directly perturb steroidogenesis. Second, spawning pairs (0.41, 4.1, 41 µg/L) (1.0, 10, 100 for 23 days assess impacts on reproduction. Lastly, male µg/L metformin, mixture both compounds, samples collected over 96-h time course hepatic transcriptome metabolome. Neither nor affected steroid production by ex vivo. Hepatic metabolome changes chemical- time-dependent, largest impact exposure guanylurea/L. Overall, did not elicit effects consistent environmentally relevant concentrations.
