The rise of antimicrobial resistance (AMR) in bacterial pathogens such as Klebsiella pneumoniae (Kp) is a pressing public health and economic concern. 'One-Health' framework recognizes that effective management AMR requires surveillance agricultural well clinical settings, particularly low-resource regions Pakistan. Here, we use whole-genome sequencing to characterise 49 isolates Klebisella spp. (including 43 Kp) 2 presumptive Providencia rettgeri recovered from dairy farms located near 3 cities Pakistan-Quetta (n = 29), Faisalabad 19), Sargodha 3). Kp corresponded 38 sequence types (STs), 35 these STs were only observed once. This high diversity indicates frequent admixture limited clonal spread on local scales. Of the isolates, 41 (84%) did not contain any clinically relevant genes (ARGs), detect ARGs predicted encode carbapenems or colistin. However, four lineages contained multiple ARGs: ST11 2), ST1391-1LV 1), ST995 1) ST985 1). 11, 1391-1LV 995 shared core set five ARGs, including blaCTX-M-15, harboured different plasmids. carried 16 genes, blaCTX-M-55. two P. also ARGs. Finally, most common plasmids which harbour our dataset non-randomly distributed between regions, suggesting expansion occurs independently host lineage. Evidence regarding how contribute emergence Pakistan valuable for authorities organizations responsible health, agriculture environment, industrial development.

A variety of methods have been developed to detect antimicrobial resistance (AMR) in different environments better understand the evolution and dissemination this public health threat. Comparisons results generated using AMR detection methods, such as quantitative PCR (qPCR) whole-genome sequencing (WGS), are often imperfect, few studies analysed samples parallel evaluate differences. In study, we compared bacterial culture WGS a culture-independent commercially available qPCR assay concordance between utility each answering research questions regarding presence epidemiology wild bird habitats. We first assessed gene 45 isolates from which had existing data. We then 52 faecal 9 spatiotemporally collected water phenotypically resistant indicator isolates.Overall was strong isolates, although differed among antibiotic classes.
