The novel brominated flame retardant, 1,2-bis(2,4,6-tribromophenoxy)ethane (BTBPE), has increasingly been detected in environmental and biota samples. However, limited information is available regarding its toxicity, especially at environmentally relevant concentrations. In the present study, adult male zebrafish were exposed to varying concentrations of BTBPE (0, 0.01, 0.1, 1, 10 μg/L) for 28 days. results demonstrated underperformance mating behavior reproductive success when paired with unexposed females. Additionally, a decline sperm quality was confirmed BTBPE-exposed zebrafish, characterized by decreased total motility, progressive increased morphological malformations. To elucidate underlying mechanism, an integrated proteomic phosphoproteomic analysis performed, revealing predominant impact on mitochondrial functions protein level universal response across different cellular compartments phosphorylation level. Ultrastructural damage, expression apoptosis-inducing factor, disordered respiratory chain involvement impairment testes. These findings not only provide valuable insights future evaluations potential risks posed similar chemicals but also underscore need further research into dysfunction health.

Legacy brominated flame retardants have been recognized as risky factors leading to declined sperm quality. The widespread utilization of decabromodiphenyl ethane (DBDPE) a replacement for ether has given rise considerable concern over its potential risks reproductive health. objectives were quickly determine whether DBDPE affects quality upon ex vivo exposure, reveal the outcomes and underlying molecular mechanisms using an in zebrafish model exposed DBDPE, validate impact on DNA damage energy metabolism balance vitro. Zebrafish spermatozoa treated with (0.01, 0.1, 1, 10μM) 3 h, motility fertilization ability normal eggs evaluated. Then adult male (0.1, 10, 100 nM) 2 months, their performance was examined. Four-dimensional label-free proteome phosphoproteome performed testes, findings validated by multiple indicators. Finally, mouse spermatogonial GC-1 cells 1μM) 72 examined, well production glycolysis oxidative phosphorylation. Ex exposure caused lower rates spermatozoa. In abnormal spermatogenesis testes. Integrated whole-proteome analysis revealed responses metabolic disorders A dosage window characterized higher mitochondrial membrane (MMP) combination unchanged reactive oxygen species apoptosis observed both testes cells. This study suggests that zebrafish, could impair spermatogenesis, mechanism be related reprogramming testicular germ https://doi.org/10.1289/EHP14426.
