Abstract Perfluoroalkyl and polyfluoroalkyl substances (PFASs) may have a role in impaired health. However, the data on association between PFASs Systemic lupus erythematosus (SLE) been limited. We designed population-based case–control study China evaluated association. 100 normal persons (Control) SLE patients (Case) were obtained from 113 controls 125 cases according to matching conditions. Serum samples collected by venipuncture for UHPLC-MRM-MS Analysis obtain concentration of five participants. Demographic characterization description was performed two groups participants, distribution described compared, then divided into three tiers (< 50th, 50th ~ 75th, > 75th) subsequent analysis. Conditional logistic regression models utilized calculate odds ratios (ORs) 95% CIs SLE. Relationship changes risk assessed restricted cubic spline. As highest serum levels tested this population, perfluoroundecanoic acid (PFUnA) quartile had 2.78-fold (95%CI: 1.270, 6.10) compared with lowest PFUnA exposure, other types also showed high as well mixture. Additionally, exposure exist dose–response relationship (ptrend < 0.05). This remained be found after adjusting covariates model 1 (adjustment BMI) 2(adjustment BMI, smoking, drinking, hypertension leukocyte). The spline illustrated gradual increase possible increasing components levels. Our firstly revealed that are factors exposures associated dose − response manner. Evidence larger more adequately powered cohort studies is needed confirm our results.

Published: Feb. 23, 2024

Abstract Background Exposure to heavy metals, particularly lead and mercury, has been identified as a significant risk factor for renal dysfunction, often through mechanisms involving oxidative stress. Despite extensive research, the specific role of serum α-klotho in modulating effects these metals on kidney function remains unclear. This study aims elucidate relationship between levels lead, function, investigate stress's potential modulatory effects, explore mediating this context among US adults, derived from National Health Nutrition Examination Survey (NHANES) data spanning 2007 2016. Methods In cross-sectional analysis 11,032 adults aged 40 79 years NHANES, we measured mercury via inductively coupled plasma mass spectrometry using enzyme-linked immunosorbent assay (ELISA). Kidney was evaluated creatinine-based estimated glomerular filtration rate (eGFR). Multivariable linear regression models were applied correlations metal concentrations, α-klotho, eGFR. Additionally, mediation model employed determine exposure function. Results Our findings indicate association reduced eGFR, suggesting impaired with increased exposure. Mediation revealed that mediated 6.10–9.75% effect highlighting its pathway Subgroup further specified significantly mediates women individuals 40–69, whereas did not show distinct patterns across gender age groups. Conclusions The demonstrates are negatively correlated playing association. These underscore importance considering health, offering new insights into preventive therapeutic impairment. Further research is warranted clinical applications targeting mitigating adverse kidneys.
