The chronic effects of triclosan (TCS) and 17α-ethinylestradiol (EE2) were assessed individually in Ceriodaphnia dubia. joint effect dubia was determined using the concentration addition (CA) independent action (IA) mathematical models. synergy antagonism resulting from binary mixtures these compounds, deviations related to dose-level dose-ratio dependence investigated. Both compounds are endocrine disruptors, present hygiene pharmaceutical products, potentially causing environmental wildlife issues. results bioassay with demonstrated that TCS an EC50 (median effective concentration) = 93 ± 1.41µg L-1 more toxic than EE2 589 2.12 µg for microcrustaceans. CA model better predicted mixture toxicity. revealed interactions between mainly showing synergism, but also dose level synergism at low levels high levels. identified this study underscore significance enhancing chemical screenings samples toxicity data, especially when dealing combinations whose not well understood.

Fatty acid synthase (FASN) has been shown to be critical in the replication of several viruses genus Orthoflavivirus. In this study role two inhibitors FASN that work through different mechanisms were investigated dengue virus (DENV) and Zika (ZIKV) infections. Triclosan is a inhibitor targets enol reductase domain FASN, while lapatinib exerts an effect on acting HER2, upstream regulator FASN. After determining cytotoxicity, comprehensive analysis these drugs DENV 2 ZIKV infection was undertaken. The results showed triclosan had moderate antiviral activity against both (EC50 = 10.21 µM; Selective index (SI) 3.99) ( EC50 22.84 SI 5.49). Lapatinib reasonable 4.9 26.09), but computer modeling suggested potential directly by binding NS5. result better fit with NS5 than NS5, confirmed as for towards µM calculated 37.92. are consistent other studies use target domains suggesting simply targeting enzymatic insufficient therapeutic drug development, lapatinib, or similar molecules may have real potential.

The effects of triclosan (TCS) and its transformation product methyl-triclosan (MTCS) on the earthworm Eisenia fetida were investigated using Gas chromatography-mass spectrometry (GC-MS) metabolomics. Earthworms exposed to 0, 0.25, 1, 4, 16, 64 µg g-1 either TCS or MTCS for 14 days in commercial worm bedding. Only exposure caused any metabolites targeted this study. Succinic acid was elevated relative control at concentrations ≥ 0.25 glucose 1 g-1. had greatest separation from Principal Components Analysis (PCA), while largest Discriminant (DA). Essential amino acids valine, leucine, serine, phenylalanine large, standardized coefficients PCA DA reflecting their contribution separation, suggesting that essential homeostasis perturbed by MTCS. Malic acid, succinic margaric highly correlated (p < 0.01) with Canonical PC 2, maltose 2 7, indicating a strong relationship between these multivariate expressed canonical variables. Disruption succinate metabolism membrane destabilization mitochondria hypothesized as possible mode action
