Ubiquitin-specific peptidase 18 (USP18) is a specific interferon-stimulated gene 15 demodifying enzyme that plays an important role in apoptosis. In this study, we investigated the of USP18 apoptosis hepatocellular carcinoma cells, especially its ability to regulate through endoplasmic reticulum (ER) stress. We found protein levels Bcl-2-associated x and cytochrome c were down-regulated by USP18, which suppressed classical mitochondrial-mediated pathway. also inhibited unfolded response (UPR) pathway inhibiting phosphorylation kinase RNA-like (PERK) expression CCAAT/enhance-binding homologous protein, downstream marker molecule ER The UPR triggered stress eventually led cleavage effecter proteases, including caspase-3, leading Furthermore, combined with PERK agonist regulated PERK-eukaryotic initiation factor-2α-activating transcription factor 4 axis UPR. Our results show participates regulation cell different pathways, pathway, it complex responses regulation.

Colorectal cancer (CRC) is the most common malignant tumor and one of lethal tumors in world. Despite treatment with a combination surgery, radiotherapy, and/or systemic treatment, including chemotherapy targeted therapy, prognosis patients advanced CRC remains poor. Therefore, there an urgent need to explore novel therapeutic strategies targets for CRC. MicroRNAs (miRNAs/miRs) are class short noncoding RNAs (approximately 22 nucleotides) involved posttranscriptional gene expression regulation. The dysregulation its recognized as key regulator related development, progression metastasis In recent years, number miRNAs have been identified regulators drug resistance CRC, some gained attention potential overcome this review, we introduce diverse mechanisms summarize therapies based on miRNAs.
