Urine analysis data obtained using modern microbiological methods and 16S rRNA gene sequencing technology indicate that the urinary system has its own microbial ecosystem. Individual microbiota members can play a key role in development of cancer. Certain bacterial taxa have been revealed bladder urothelial carcinoma cells affect carcinogenesis, treatment response, relapses through various mechanisms. The studies are conducted to use not only vaccine strains, but also probiotic strains oncolytic bacteria for prevention relapses.

Abstract The human body harbors a diverse ecosystem of microorganisms, including bacteria, viruses, and fungi, collectively known as the microbiota. Current research is increasingly focusing on potential association between microbiota various neuropsychiatric disorders. resides in parts body, such oral cavity, nasal passages, lungs, gut, skin, bladder, vagina. gut gastrointestinal tract has received particular attention due to its high abundance role psychiatric neurodegenerative However, presents other tissues, though less abundant, also plays crucial immune system homeostasis, thus influencing development progression For example, imbalance associated periodontitis might increase risk for Additionally, studies using postmortem brain samples have detected widespread presence bacteria brains patients with Alzheimer’s disease. This article provides an overview emerging host disorders discusses future directions, underlying biological mechanisms, reliable biomarkers microbiota, microbiota-targeted interventions, this field.

Abstract Background Through research on the gut microbiota (GM), increasing evidence has indicated that GM is associated with esophageal cancer (ESCA). However, specific cause-and-effect relationship remains unclear. In this study, Mendelian randomization (MR) analysis was applied to investigate causal between and ESCA, including its subtypes. Methods We collected information 211 GMs acquired data ESCA subtypes through genome-wide association studies (GWASs). The primarily assessed using inverse variance weighted (IVW) method. Additionally, we median estimator (WME) method, MR–Egger mode, simple mode provide further assistance. Subsequent these analyses, sensitivity conducted intercept test, MR-PRESSO global leave-one-out Result Following our assessment five methods analysis, identified seven potential relationships At genus level, Veillonella Coprobacter were positively correlated whereas Prevotella9 , Eubacterium oxidoreducens group Turicibacter negatively ESCA. case of adenocarcinoma (EAC), Flavonifractor exhibited a positive correlation, while Actinomyces negative correlation. Conclusion Our study revealed subtypes, offering novel insights for advancement diagnosis treatment.

Background: Increasing evidence points to the urinary microbiota as a possible key susceptibility factor for early-stage bladder cancer (BCa) progression. However, interpretation of its underlying mechanism is often insufficient, given that various environmental conditions have affected composition microbiota. Herein, we sought rule out confounding factors and clarify how Eubacterium sp. CAG:581 promoted non-muscle invasive (NMIBC) development. Methods: Differentially abundant 51 NMIBC patients 47 healthy controls (as Cohort 1) were first determined by metagenomics analysis. Then, modeled coculture organoids with candidate in anaerobic explored differentially expressed genes these tissues RNA-Seq. Furthermore, dissected mechanisms involved into inducing extracellular matrix protein 1 (ECM1) metalloproteinase 9 (MMP9) upregulation. Finally, used multivariate Cox modeling investigate clinical relevance 16S ribosomal RNA (16SrRNA) levels prognosis 406 2). Results: infection accelerated proliferation (p < 0.01); ECM1 MMP9 most upregulated induced increased colony forming units (CFU) gradient via phosphorylating ERK1/2 1. Excluding favorable impact potential contributing factors, ROC curve 2 manifested 3-year AUC value 0.79 cut-off point 16SrRNA 10.3 (delta CT value). Conclusion: Our suggests progression through ECM1/MMP9 pathway, which may serve promising noninvasive diagnostic biomarker NMIBC.

Microorganisms have been identified in tumor specimens for over a century. It is only recent years that tumor-associated microbiota has become rapidly expanding field. Assessment techniques encompass methods at the frontiers of molecular biology, microbiology, and histology, requiring transdisciplinary process to carefully decipher this new component microenvironment. Due low biomass, study poses technical, analytical, biological, clinical challenges must be approached as whole. To date, several studies begun shed light on composition, functions, relevance microbiota. This piece microenvironment puzzle could potentially change way we think about treat patients with cancer.

SUMMARYBacillus Calmette-Guérin (BCG) has been the standard treatment for non-muscle-invasive bladder cancer over 30 years. Despite its proven efficacy, challenges persist, including unclear mechanisms of action, resistance in 30%-50% patients, and significant side effects. This review presents an updated balanced discussion antitumor BCG, focusing on direct effects interactions with various cell types within tumor microenvironment. Notably, recent research between BCG microbiome is also incorporated. We further summarize analyze latest preclinical clinical studies regarding both intrinsic adaptive to cancer. Based current understanding BCG's therapeutic principles mechanisms, we systematically explore strategies improve BCG-based immunotherapy. These include development recombinant combination therapy different drugs, optimization regimens management, exploration new approaches by targeting changes microbiota metabolites. measures aim effectively address cancer, reduce toxicity, ultimately enhance anti-tumor efficacy. Bacterial therapy, represented genetically engineered oncolytic bacteria, gradually emerged field As only bacterial drug successfully approved oncology use, provided decades experience. By consolidating lessons from successes limitations, hope provide valuable insights application therapies treatment.

Microbiomes have been linked to oncogenesis, e.g. the intestinal microbiome and colon cancer or HPV-associated cervical cancer. A connection between microbiomes of different body cavities tumor oncogenesis was shown. The gut microbiome's influence on bladder established, raising question whether nearby (rectum, vagina) also due their proximity. Considering various broader microbial components, this systematic review aims investigate differences in bladder, vaginal, microbiota-including bacterial, viral, fungal archaea-between patients with healthy controls. Databases (PubMed, Scopus, Embase) were searched until April 2022. Three types studies included: "(1) using control groups (case-controlled studies) (2) that provided information presence abundance taxa (3) increased decreased and/or groups.". Risk bias assessed Newcastle Ottawa Scale. Fourteen (695 samples: 403 cancer, 292 controls) analyzed.
