The role of peroxiredoxin 1 (PRDX1), a crucial enzyme that reduces reactive oxygen and nitrogen species levels in HepG2 human hepatocellular carcinoma (HCC) cells, the regulation HCC cell stemness under oxidative stress underlying mechanisms remain largely unexplored. Here, we investigated therapeutic potential non-thermal plasma targeting cancer stem cells (CSCs) HCC, focusing on resistance to PRDX1. By simulating conditions using plasma-activated medium, found reduction PRDX1 resulted considerable increase apoptosis, suggesting plays key defense CSCs. Furthermore, had higher spheroid formation capability increased markers (CD133, c-Myc, OCT-4), indicating strong stemness. Interestingly, expression was notably than other types such as Hep3B Huh7 whereas PRDX family proteins (PRDX 2–6) were relatively consistent. inhibition peroxidase activity by conoidin A markedly reduced traits death rate. xenograft mouse model, downregulation considerably inhibited solid tumors after medium (PAM) treatment. These findings underscore critical regulating apoptosis stress, highlighting promising target for NTP-based treatment HCC.

Accumulating evidence has demonstrated that F-box protein 22 (FBXO22) participates in tumour development and progression various types of human malignancies. However, the functions detailed molecular mechanisms FBXO22 osteosarcoma tumorigenesis remain elusive. In this study, we aimed to determine effects on cell proliferation, migration invasion cells using counting kit-8 Matrigel Transwell approaches. Moreover, explored by which mediated oncogenesis via Western blotting, immunoprecipitation ubiquitination. We found depletion inhibited cells, whereas overexpression increased proliferation motility cells. Mechanistically, promoted ubiquitination degradation FoxO1 reduced regulation FoxO1. Taken together, our findings provide new insight into FBXO22-induced tumorigenesis. The inhibition could be a promising strategy for treatment osteosarcoma.

Abstract Glioma, a common malignancy, is characterized by high morbidity and mortality. Promoting ferroptosis can delay tumor progression. Here, we aimed to explore the underlying mechanism of in glioma. In vitro vivo experiments were conducted using glioma cells nude mice. The expression genes proteins was evaluated RT‐qPCR, Western blot assay, immunohistochemical staining. Malignant activities MTT, EdU, Transwell assays. levels Fe 2+ , lipid reactive oxygen species, malondialdehyde determined commercial kits. interplays among CMTM5, WWP2, LATS2 validated Co‐immunoprecipitation assay. UALCAN database predicted downregulation CMTM5 glioma, low associated with poor survival outcomes. overexpression inhibited cell growth invasion promoted cells. Besides, protein interacted WWP2 decreased expression. silencing attenuated ubiquitination enhance phosphorylation YAP1. exerted suppressive effect on regulating WWP2/LATS2 pathway. experiments, suppressed enhanced ferroptosis. regulated Hippo/YAP signaling inhibit promote WWP2‐mediated ubiquitination, thereby attenuating

Protein post-translational modifications (PTMs) represent a crucial aspect of cellular regulation, occurring after protein synthesis from mRNA. These modifications, which include phosphorylation, ubiquitination, acetylation, methylation, glycosylation, Sumoylation, and palmitoylation, play pivotal roles in modulating function. PTMs influence localization, stability, interactions, thereby orchestrating variety processes response to internal external stimuli. Dysregulation is linked spectrum diseases, such as cancer, inflammatory neurodegenerative disorders. UFMylation, type PTMs, has recently gained prominence for its regulatory role numerous processes, including stress, key signaling pathways influencing functions. This review highlights the function UFMylation development progression tumors, underscoring potential therapeutic target. Moreover, we discuss tumorigenesis malignant progression, explore impact on cancer immunotherapy. The article aims provide comprehensive overview biological functions propose how targeting could enhance effectiveness immunotherapy strategies.
