Over the last few years, development of fluorescent probes has received considerable attention. Fluorescence signaling allows noninvasive and harmless real-time imaging with great spectral resolution in living objects, which is extremely useful for modern biomedical applications. This review presents basic photophysical principles strategies rational design as visualization agents medical diagnosis drug delivery systems. Common phenomena, such Intramolecular Charge Transfer (ICT), Twisted (TICT), Photoinduced Electron (PET), Excited-State Proton (ESIPT), Fluorescent Resonance Energy (FRET), Aggregation-Induced Emission (AIE), are described platforms fluorescence sensing vivo vitro. Probes allowing high-contrast discrimination of cancer cells and effective retention are powerful tools for the early diagnosis treatment cancer. However, conventional small-molecule probes often show limited performance in both aspects. Herein, we report an ingenious molecular engineering strategy tuning cellular uptake rhodamine dyes. Introduction polar aminoethyl leads to increased brightness reduced dyes, this change can be reversed by amino acetylation. Moreover, these modifications allow take up more dyes than normal (16-fold) through active transport. Specifically, further improve signal contrast (56-fold) between constructing activatable confirm that released fluorophore remain with extended time, enabling long-term specific tumor imaging.
