Abstract Developing single fluorescent probe for simultaneously visualizing mitophagy flux and oxidative stress with super‐resolution is highly demanded yet quite challenging. Herein, a ratiometric HClO AHOH designed synthesized which capable of staining lysosomes mitochondria red green colour, respectively. could be selectivity oxidized by HClO, leading to large emission blue shift (90 nm) an over 1300‐fold enhancement the ratio Fl 547nm / 637nm . We apply in microscopy clearly visualize dynamics mitochondria–lysosomes interactions states upon different stimuli. Mitochondria dysfunction triggered drugs genetic defect lead elevated higher levels mitophagy. Moreover, serve as reliable tool evaluating efficacy regulating dysfunction. This work provides powerful dual‐colour imaging agent real‐time monitoring organelle stress.
