Abstract Emotional states influence bodily physiology, as exemplified in the top-down process by which anxiety causes faster beating of heart 1–3 . Abstract Interoception, the ability to timely and precisely sense changes inside body, is critical for survival 1–4 . Vagal sensory neurons (VSNs) form an important body-to-brain connection, navigating visceral organs along rostral–caudal axis of body crossing surface–lumen into appropriate tissue layers 5,6 The brain can discriminate numerous signals through VSNs, but underlying coding strategy remains poorly understood. Here we show that VSNs code organ, layer stimulus modality—three key features interoceptive signal—in different dimensions. Large-scale single-cell profiling from seven major in mice using multiplexed projection barcodes reveals a ‘visceral organ’ dimension composed differentially expressed gene modules body’s axis. The inadequacy of a categorial approach to mental health diagnosis is now well-recognised, with many authors, diagnostic manuals and funding bodies advocating dimensional, trans-diagnostic research. Interoception, the perception of internal bodily states, is thought to be inextricably linked affective qualities such as anxiety. Although interoception spans sensory metacognitive processing, it not clear whether anxiety differentially related these processing levels. Here we investigated this question in domain breathing, using computational modeling and high-field (7 T) fMRI assess brain activity relating dynamic changes inspiratory resistance varying predictability. Notably, anterior insula was associated with both breathing-related prediction certainty errors, suggesting an important role representing updating models body. Individuals low versus moderate traits showed differential for certainty. Interoception - the physiological sense of our inner bodies has risen to forefront psychological and psychiatric research. Much this research utilizes tasks that attempt measure ability accurately detect cardiac signals. Unfortunately, these approaches are confounded by well-known issues limiting their validity interpretation. At core controversy is role subjective beliefs about heart rate in confounding measures interoceptive accuracy. Here, we recast as an important part causal machinery interoception, offer a novel psychophysical "heart discrimination" method estimate accuracy precision. By applying task 223 healthy participants, demonstrate more biased, less precise, associated with poorer metacognitive insight relative exteroceptive control condition.
