The bidirectional regulation between the gut microbiota and brain, known as gut-brain axis, has received significant attention. myelin sheath, produced by oligodendrocytes or Schwann cells, is essential for efficient nervous signal transmission maintenance of brain function. Growing evidence shows that both oligodendrogenesis myelination are modulated its metabolites, when dysbiosis occurs, changes in composition and/or associated metabolites may impact developmental occurrence neurodevelopmental disabilities. Although link demyelinating disease such multiple sclerosis been extensively studied, our knowledge about role other myelin-related disorders, neurodegenerative diseases, limited. Mechanistically, microbiota-oligodendrocyte axis primarily mediated factors inflammation, vagus nerve, endocrine hormones, evidenced metagenomics, metabolomics, vagotomy, morphological molecular approaches. Treatments targeting this include probiotics, prebiotics, microbial herbal bioactive compounds, specific dietary management. In addition to commonly used approaches, viral vector-mediated tracing gene manipulation, integrated multiomics multicenter clinical trials will greatly promote mechanistic interventional studies ultimately, development new preventive therapeutic strategies against gut-oligodendrocyte axis-mediated impairments. Interestingly, recent findings showed can be induced hippocampal damage reversible myelin-targeted drugs, which provides insights into understanding how hippocampus-based functional impairment (such Alzheimer's disease) regulates peripheral homeostasis systemic disorders.

ABSTRACT Parkinson's disease (PD) is a prevalent neurodegenerative caused by the death of dopaminergic neurons within substantia nigra pars compacta (SNpc) region midbrain. Recent genomic and single cell sequencing data identified oligodendrocytes oligodendrocyte precursor cells (OPCs) to confer genetic risk in PD, but their biological role unknown. Although SNpc are scarcely or thinly myelinated, there gap knowledge concerning physiological interactions between oligodendroglia. We sought investigate distribution OPCs with regard myelination state mouse (SN) high‐resolution imaging provide morphological assessment OPC‐dopaminergic neuron quantification numbers across different age groups. evenly distributed midbrain throughout lifespan they physically interact both soma axons neurons. Spinocerebellar ataxia type 3 (SCA3) is the most common dominantly inherited and belongs to family of nine diseases caused by a polyglutamine expansion in disease-causing protein. In SCA3, ATXN3 causes neuron loss disease-vulnerable brain regions, resulting progressive coordination ultimately death. There are no disease-modifying or preventative treatments for this uniformly fatal disorder. Recent studies demonstrate prominent white matter atrophy microstructural alterations regions SCA3 patients mouse models. Spinocerebellar ataxia type 3 (SCA3), a neurodegenerative disorder caused by excess CAG repeats in the ATXN3 gene, leads to progressive cerebellar and other symptoms. The results of previous studies suggest that trace element dysregulation contributes onset. Here, we investigated relationships with repeat length, clinical severity, brain structural functional connectivity 45 patients SCA3 44 healthy controls (HCs). Blood levels lithium (Li), selenium (Se), copper (Cu) were significantly lower than HCs; Li Se negatively correlated especially manifest subgroup. Diffusion tensor imaging combined resting-state magnetic resonance revealed fractional anisotropy white matter (WM) bilateral frontal parietal regions; tractography mapping showed Li-associated region nerve fiber pathways SCA3. Dynamic causal modeling analyses bidirectional from inferior lobule(IPL) cerebellum was blood level Time series correlation-based analysis intrinsic connectivities dorsal premotor cortex(PMd) IPL local regions weaker HCs. Our dysregulation, deficiency, induces alterations manifestations SCA3; supplementation may be beneficial for WM or astrocytes this patient population.

While oligodendrocytes are able undergo endogenous repair process known remyelination, this becomes inefficient usually fails in presence sustained inflammation. The present review focuses our current knowledge regarding activation innate adaptive systems chronic demyelinating disease, multiple sclerosis, provides other conditions, such perinatal white matter injury, traumatic brain viral infections, converges injury.
