Drug toxicity and related drug-induced liver injury (DILI) have become major biosafety issues. Enzyme-activated fluorescent probes been demonstrated as a powerful tool for the diagnosis of DILI; however, they suffer from diffusive signal dilution interference with other organs, thus leading to misassessment drug inaccurate diagnosis. Alkaline phosphatase (ALP) is an important biomarker, alterations in enzyme level are tightly correlated severity damage. Herein, enzyme-activated intramolecular hydrogen bond (IMHB) enhanced probe (TPEG-P) was developed ALP detection cells mice models DILI. Differing previously reported intermolecular charge transfer (ICT) or excited-state proton (ESIPT) mechanisms, TPEG-P enables precise recognition imaging only through activation bonds could situ sensitively detect varying degrees caused by toxicity. This IMHB strategy will advance development bioimaging future.

In this work, we first designed and synthesized tetraphenylene-fused aryl-imidazole derivatives TM-1-4 via regulation of molecular structure, which were consisted 1H-imidazo[4,5-f][1,10]phenanthroline, 1H-phenanthro[9,10-d]imidazole, 4,5-diphenyl-1H-imidazole, 3,3'-(1H-imidazole-4,5-diyl)dipyridine moieties AIE-active tetraphenylethene units, respectively. The results illustrated that exhibited clear AIE characteristics. Meanwhile, TM-2 TM-3 show excellent solid emission properties (ΦTM-2 =13.73 % ΦTM-3 =36.21 %), whereas TM-1 TM-4 exhibit the opposite (ΦTM-1 =1.48 ΦTM-4 =4.83 %). multiple rotors (pyridine benzene ring) causes twisted conformations molecule prevents π-π stacking enhances emission(ΦTM-2<ΦTM-3, ΦTM-1<ΦTM-4). Significantly, also reversible mechanochromic behavior (Emission red shifts: ΔλTM-2 =43 nm ΔλTM-3 =41 nm) with color changes between blue green emissions. powder X-ray diffraction (PXRD) suggested disordered state ground sample could be readily returned to an ordered crystalline. Therefore, mechanochromisms are ascribable phase transformation crystal amorphous structure. single analysis reveals a conformation for TPE moiety absence intermolecular stacking. These optical make them potentially applications in materials imaging agents.
