Smart-responsive materials have attracted much attention in the enrichment of post-translational modifications proteins. In this work, for first time, we developed a smart strategy (MNPs-l-DOPA/PEI-SP) based on change hydrophilic properties spiropyran under regulation light and pH to realize controllable release intact glycopeptides. The mechanism possible binding were verified by theoretical calculations. platform MNPs-l-DOPA/PEI-SP was used screen glycoprotein biomarkers hepatocellular carcinoma (HCC) evaluate its cancer diagnostic monitoring performance. A total 3,864 N-glycopeptides containing 166 N-glycoproteins successfully identified serum samples early-stage HCC patients, while 3,266 193 glycoproteins normal control (NC) samples. This work not only provides new ideas efficient glycopeptides with smart-responsive material, but also broadens research possibilities biomarker discovery liquid biopsies.

The comprehensive investigation of protein phosphorylation and glycosylation aids in the discovery novel biomarkers as well understanding pathophysiology illness. In this work, a nitrogen/titanium-rich porous organic polymer was developed by copolymerizing carbohydrazide (CH) 2,3-dihydroxyterephthalaldehyde (2,3-Dha) modifying with Ti4+ (CH-Dha-Ti4+). adequate nitrogen contributes to enrichment glycopeptides via HILIC, while titanium benefits from capturing phosphopeptides through IMAC. proposed method exhibits excellent selectivity (1 : 1000, both for phosphopeptides), LOD (for glycopeptides: 0.05 fmol μL-1, phosphopeptides: 0.2 fmol), loading capacity 100 mg g-1, 125 g-1) size-exclusion effect 10 000, phosphopeptides). Furthermore, CH-Dha-Ti4+ applied capture human serum; 205 45 were detected serum normal controls; 294 63 found uremia patients after being analyzed nano LC-MS/MS. discovered involved several molecular biological processes activities, according gene ontology study.
