The development of reliable probes for in vivo detection hydrogen sulfide (H2S) with high sensitivity and selectivity is great significance due to its key roles many pathological physiological processes. Herein, it was found that H2S could finely regulate surface structure gold nanoclusters (AuNCs) through reduction Au(I)-ligand motifs further quench their fluorescence by a two-stage kinetic reaction process. Stage I showed the H2S-assisted Au(0) core growth rapid decrease; stage II optimization reconstruction relatively slow quenching. By virtue excellent response AuNCs H2S, novel ratiometric probe (RBDA) sensing designed electrostatic attraction-induced resonance energy transfer (FRET) between rhodamine B. facilely prepared, showing straightforward, built-in self-calibration. It presented limit (LOD) 56 nM an over various other biological species. demonstrated possess biostability, low cytotoxicity, good cell issue penetrability, favorable biocompatibility. realizes successful monitoring both exogenous endogenous levels living cells zebrafish.

Rheumatoid arthritis (RA), a common chronic inflammatory illness, is still incurable, reducing the sufferers' quality of life significantly. Adenosine 5′-triphosphate (ATP) and hypochlorous acid (HOCl) are key indicators in RA, but their precise mechanisms RA pathophysiology unknown. As result, order to detect ATP HOCl simultaneously, we created two new dual-channel/localization single-molecule fluorescence probes, RhTNMB RhFNMB. Furthermore, RhFNMB outperformed terms detection performance. produce independent responses light red channel (λex = 520 nm, λem 586 nm) deep 620 688 nm), respectively, without spectral crosstalk. It should be noted that probe successfully imaged mitochondria cells. Surprisingly, demonstrated remarkable ability diagnosis treatment Pseudomonas aeruginosa-induced abdominal inflammation mice. We continued apply track discovered concentrations were considerably greater joints than normal joints. also confirmed therapeutic effect methotrexate on RA. This study first achieve dual-channel imaging HOCl, which great value for early therapy
