Here, a novel rapid and ultrasensitive aptamer biosensor was designed for target-induced activation of AIE effect followed by the Crispr Cas12a (LbCpf1)-mediated cleavage to achieve dual-signal detection. The prepared DNA building blocks contain target aptamer, ssDNA-Fc, Activator1. In this system, mode divided into two steps. First, when interacts with aptamers, would be disintegrated rapidly, releasing mass Ac1, generating ETTC-dsDNA aggregated produce fluorescence signal effect. Second, release Ac2, LbCpf1-crRNA activated, which greatly improves ssDNA-Fc efficiency render amplification detection target. Satisfactorily, using approach detect gliotoxin, optimal conditions achieved reducing time 55 min, achieving low limit 2.4 fM satisfactory linear in range 50 1 nM, addressed shortcoming weak electrochemical previous sensors. water-insoluble material coupled obtain water-soluble successfully introduced sensor 5.6 fM.
