The worldwide spread of antibiotic resistance is considered to be one the major health threats society. While developing new antibiotics crucial, there also a strong need for next-generation analytical methods studying physiological state live bacteria in heterogeneous populations and their response environmental stress. Here we report single-cell high-throughput method monitor changes bacterial cell envelope stress based on ratiometric flow cytometry. We used sensitive fluorescent molecular probe, Nile Red-based solvatochromic antimicrobial peptide UNR-1, with defined cellular localization Gram-positive Gram-negative bacteria. developed robust protocol calculating generalized polarization (GP) fluorescence adapted Our methodology enabled rapid detection perturbations caused by exposure antibiotics, heat shock, other factors.

Summary Plasma membrane (PM) nanodomains have emerged as pivotal elements in the regulation of plant cellular functions and signal transduction. These nanoscale regions, enriched specific lipids proteins, behave regulatory/signaling hubs spatially temporally coordinating critical functions. In this review, we first examine mechanisms underlying formation maintenance PM cells, highlighting roles lipid composition, protein oligomerization interactions with cytoskeletal cell wall components. Then, discuss how act organizing centers by mediating protein–protein that orchestrate essential processes such symbiosis, defense against pathogens, ion transport or hormonal reactive oxygen species (ROS) signaling. Finally, introduce concept nanoenvironments, where localized physicochemical variations are generated very close proximity nanodomains, response to stimuli. After decoding a dedicated machinery likely vicinity enrichment secondary messengers, ROS Ca 2+ , would allow downstream responses. This review provides insights into dynamic nature proposes future research better understand their contribution intricate signaling networks govern development stress
