Herein, we report a naturally occurring flavonol, quercetagetin 5,6,7,3′,4′-pentamethyl ether (known as marionol), nontoxic fluorescent probe for rapid, sensitive, and selective detection of Cu2+ in water. The interaction between marionol ions is studied by various state-of-the-art spectroscopic techniques such 1H NMR, UV–vis, fluorescence, high-resolution mass spectrometry (HRMS). Marionol shows strong fluorescence quenching the presence Cu2+, whereas other interfering cations Fe3+, Co2+, Al3+, Pb2+, Hg2+, Zn2+, Mg2+, Mn2+, Ca2+, Cu+, Ag+, Na+, K+ produce negligible changes fluorescence. binding stoichiometry found to be 2:1, according Job's plot HRMS analyses. constant (Ka) with calculated 2.89 × 109 M–2, limit 0.15 μM. In addition, further used quantification natural spring water samples. good recoveries (92–110% RSD <5%) its low toxicity MCF-7 human breast cancer cells make unique plant-based ions.

Metal ions are essential for all life. In microbial cells, potassium (K+) is the most abundant cation and plays a key role in maintaining osmotic balance. Magnesium (Mg2+) dominant divalent required nucleic acid structure as an enzyme cofactor. Microbes typically require transition metals manganese (Mn), iron (Fe), copper (Cu), zinc (Zn), although precise set of metal needed to sustain life variable. Intracellular pools can be conceptualized chemically complex mixture rapidly exchanging (labile) ions, complemented by those reservoirs that exchange slowly relative cell metabolism (sequestered). Labile buffered transient interactions with anionic metabolites macromolecules, ribosome playing major role. Sequestered include many metalloproteins, cofactors, storage depots, some redeployed upon depletion. Here, I review size, composition, dynamics intracellular highlight gaps understanding.

Copper is an essential nutrient for sustaining vital cellular processes spanning respiration, metabolism, and proliferation. However, loss of copper homeostasis, particularly misregulation loosely bound ions which are defined as the labile pool, occurs in major diseases such cancer, where tumor growth metastasis have a heightened requirement this metal. To help decipher role etiology we report histochemical activity-based sensing approach that enables systematic, high-throughput profiling status across many cell lines parallel. Coppermycin-1 reacts selectively with Cu(I) to release puromycin, then incorporated into nascent peptides during protein translation, thus leaving permanent dose-dependent marker can be visualized standard immunofluorescence assays. We showcase utility platform screening pools National Cancer Institute’s 60 (NCI-60) human line panel, identifying types elevated basal levels copper. Moreover, use show lung cancer cells activation nuclear factor-erythroid 2-related factor 2 (NRF2) possess lower resting and, result, reduced viability when treated chelator. This work establishes methods detection used assess cuproplasia, emerging form copper-dependent proliferation, providing starting point broader investigations roles transition metal signaling biology medicine.
