The comprehensive characterization of per- and polyfluoroalkyl substances (PFASs) is necessary for the effective assessment management risk at contaminated sites. While current analytical methods are capable quantitatively measuring a number specific PFASs, they do not provide complete picture thousands PFASs that utilized in commercial products potentially released into environment. These unmeasured include many PFAS precursors, which may be converted related chemicals through oxidation. total oxidizable precursor (TOP) assay offers means bridging this gap by oxidizing unknown precursors intermediates converting them stable with established standards. application TOP to samples from PFAS-contaminated sites has generated several new insights, but it also presented various technical challenges laboratories. Despite increased literature studies assay, there critical growing method beyond researchers academia. This article outlines benefits using aqueous site assessments suggests ways address some its limitations.

Per- and polyfluoroalkyl substances (PFAS) are a diverse class of highly persistent anthropogenic chemicals that detectable in the serum most humans. PFAS exposure has been associated with many adverse effects on human health including immunotoxicity, increased risk certain cancers, metabolic disruption. binding to abundant blood proteins (human albumin [HSA] globulins) is thought affect transport active sites, toxicity, elimination half-lives. However, few studies have investigated competitive these serum. Here, we use C18 solid-phase microextraction fibers measure HSA–water globulin–water distribution coefficients (DHSA/w, Dglob/w) for carbon chains containing 4 13 perfluorinated carbons (ηpfc = 4–13) several functional head-groups. ηpfc < 7 were bound HSA relative globulins, whereas ≥ showed greater propensity globulins. Experimentally measured DHSA/w Dglob/w concentrations successfully predicted variability We estimated unbound fraction varied by up factor 2.5 among individuals participating 2017–2018 U.S. National Health Nutrition Examination Survey. These results suggest globulins important covariates epidemiological aimed at understanding exposure.

Environmental contamination by fluorinated chemicals, in particular chemicals from the per- and polyfluoroalkyl substances (PFAS) class, has raised concerns around globe because of documented adverse impacts on human health, wildlife, ecosystem quality. Recent studies have indicated that pesticide products may contain a variety meet PFAS definition, including active ingredients themselves. Given pesticides are some most widely distributed pollutants across world, legacy addition into could be widespread wide-ranging implications agriculture food water contamination, as well presence rural environments.
