Inflammatory bowel diseases (IBDs) are driven by an exaggerated inflammatory response, which leads to a marked increase in oxidative stress. This, turn, exacerbates the process and causes significant cellular tissue damage. Intestinal dysbiosis, common observation IBD patients, alters production of bacterial metabolites, including short-chain fatty acids (SCFAs), key by-products dietary fiber fermentation. While role SCFAs intestinal physiology is still being elucidated, this study aimed investigate their effects on stress, particularly under conditions induced proinflammatory mediator tumor necrosis factor alpha (TNF-α). The Caco-2/TC7 cell line was employed as vitro model epithelium, cells were treated with range SCFAs, acetate, propionate, butyrate. levels protein lipid oxidation quantified, well activity antioxidant enzymes. Our findings demonstrate that microbiota-derived can effectively mitigate TNF-α-induced stress modulating enzyme activity. TNF-α induces peroxidation inhibiting catalase glutathione peroxidase activities. able upregulate restore levels. These results underscore critical gut microbiota maintaining homeostasis highlight therapeutic potential managing stress-related pathologies.

Highlights•Huaier can be an effective drug to reduce oxaliplatin-induced hepatotoxicity.•ROS-mediated oxidative stress is involved in hepatotoxicity.•Huaier relieves liver stress.•Inhibition of PI3K pathway reverses the hepatoprotective effect Huaier.AbstractHepatotoxicity caused by anticancer medication oxaliplatin (OXA) significantly restricts its clinical use and raises risk damage. Huaier, a fungus found China, has been demonstrated have various beneficial effects adjuvant therapy for cancer. However, preventive impact Huaier against OXA-induced hepatotoxicity still unknown. The potential molecular pathways behind activity were investigated current study Mice intraperitoneally injected with 10 mg/kg OXA once week six consecutive weeks establish injury model. (2 g/kg, 4 8 g/kg) was administered weekly mice gavage weeks. Commercial kits used determine contents glutathione, catalase, superoxide dismutase, malondialdehyde. Quantitative real-time polymerase chain reaction (qRT-PCR) Western blotting assess on expression pathway. exhibited good protective dose-dependent manner, which connected suppression stress, according results biochemical index detection histological staining analysis. In addition, could counteract PI3K/AKT signaling Moreover, activation eradicated LY294002. These findings imply that decreasing minimize injury, establishing groundwork lessen chemotherapy-induced practice.

This study aimed to investigate the effects of soybean bioactive peptide (SBP) on growth performance and intestinal health yellow-feathered broilers further elucidate regulatory mechanisms using multi-omics analysis. A total 320 1-day-old were randomly divided into two groups, with 10 replicates per group 16 birds replicate. Broilers in control received basal diet, those experimental (SBPG) diet 0.2 % SBP replacing same amount meal. The experiment lasted for 70 d. results showed that, compared group, final body weight average daily gain SBPG significantly higher (P < 0.05), feed conversion ratio was lower 0.05). Notably, improved gut chickens, including increased villus height, decreased levels proinflammatory factors, such as IL-1β interferon-γ, upregulated expression tight junction proteins, ZO-1 occludin. In addition, transcriptome sequencing revealed that exhibited significant enrichment multiple metabolic pathways, fatty acid metabolism, degradation, biosynthesis unsaturated acids Cecal 16S rRNA abundance butyrate-producing beneficial bacteria Muribaculaceae. Subsequent cecal metabolome analysis also enhanced lipid-related alpha-linolenic metabolism GPI-anchor biosynthesis. conclusion, is a potential additive can improve morphology, enhance immunity barrier function, optimize structure microbiota, function.
