This study aimed to explore the ameliorative effects and potential mechanisms of Huangshan Umbilicaria esculenta polysaccharide (UEP) in dextran sulfate sodium-induced acute ulcerative colitis (UC) UC secondary liver injury (SLI). Results showed that UEP could ameliorate both colon pathologic injuries, upregulate mouse intestinal tight junction proteins (TJs) MUC2 expression, reduce LPS exposure, thereby attenuating gut–liver axis. Importantly, significantly downregulated secretion levels TNF-α, IL-1β, IL-6 through inhibition NF-κB pathway activated Nrf2 signaling increase expression SOD GSH-Px. In vitro, inhibited LPS-induced phosphorylation P65 promoted nuclear translocation RAW264.7 cells. These results revealed ameliorated SLI Nrf2-mediated inflammation oxidative stress. The first investigated anticolitis effect UEP, suggesting its for treatment colitis-associated disease.
