Abstract The imbalance between osteoblasts and osteoclasts is the cause of osteoporosis. Milk-derived extracellular vesicles (mEVs), excellent drug delivery nanocarriers, can promote bone formation inhibit resorption. In this study, we conjugated bone-targeting peptide (AspSerSer, DSS)6 to mEVs by click chemistry then loaded with SRT2104, a SIRT1 (silent mating-type information regulation 2 homolog 1) agonist that was proofed help reduce loss. engineered (DSS)6-mEV-SRT2104 had intrinsic anti-osteoporosis function SRT2104 reverse in homeostasis simultaneously regulating osteogenesis osteoclastogenesis. Furthermore, labelled MnB nanoparticles be used for vivo magnetic resonance imaging (MRI) visualization. obtained nanocomposites significantly prevented loss osteoporosis mice increased mineral density, exhibiting superior accumulation under MRI. We believe proposed (DSS)6-mEV-SRT2104/MnB provides novel paradigm treatment monitoring.

Milk-derived extracellular vesicles (EVs) show potential applications as nanomedicine; however, their membrane lipids are not fully studied. Herein, we present the lipidomic analysis of extracted EVs and milk fat globules (MFGs) from human breast using ultraperformance supercritical fluid chromatography combined with quadrupole time-of-flight mass spectrometry. A total 11 lipid species, including glycerophospholipids, sphingomyelin (SM), ceramide (Cer), cholesterol (CHOL), 182 molecular species were identified quantified. in ∼4.4% polar lipids, while remaining come MFGs. Lipids rich CHOL, SM, phosphatidylcholine, phosphatidylethanolamine stable during lactational stages. In milk, MFGs have a similar CHOL/SM molar ratio (∼6). Membrane low unsaturation degree high content long-chain saturated fatty acids 24:1 n-9. The unique composition milk-derived compared to can be used study mechanism mRNA delivery infant gastrointestinal tract.

Milk-derived extracellular vesicles (MEV) have received increasing attention due to their physiological benefits for newborn growth and development. However, characteristics immunomodulatory capacities during lactation remain unclear. In this study, we isolated MEV from porcine milk at day 1, week 2, 3 after parturition compared differences by common EV characterization, showing the higher concentration smaller mean size of colostrum. Porcine could be internalized RAW264.7 macrophages functionally deliver immune-related microRNAs (miRNAs). Furthermore, promoted macrophage polarization toward M2-like phenotype exerted anti-inflammatory effects in vitro. Mice models with sepsis also displayed that suppressed secretion pro-inflammatory cytokines serum drove an spleen, ultimately weakening immune responses. Especially, colostrum exhibited most robust on immunoregulation. Overall, study provides valuable information patterns bioactivities.

Notably, 18 upregulated O-EVs, such miR-339-5p, miR-125a, miR-423-3p, miR-29c, which mainly endocytosis, focal adhesion, adherens junction. KEGG pathway further indicated that glycerophospholipid metabolism. These results provide an insight into functional roles EVs.
