Phenazine-1-carboxamide (PCN) has been exploited as a successful biopesticide due to its broad-spectrum antifungal activity. We engineered PCN-overproducing Pseudomonas chlororaphis strain through overexpressing shikimate pathway genes (aroB, aroQ, aroE, and phzC) deleting negative regulatory (relA, fleQ, pykF). The optimized produced 1.92 g/L PCN with yield of 0.11 g/g glycerol, the highest titer ever reported by using minimal media. To gain deeper insights into underlying network, final parental were examined three distinct omic data sets. 13C-metabolic flux analysis revealed substantial reconfiguration in strain, including activation EDEMP cycle, PP pathway, glyoxylate shunt, pathway. Metabolomic results indicated that central carbon was rerouted Transcriptomic identified global gene expression changes. This study forms basis for further engineering strains achieve outstanding performance.

Natural products (NPs) have attracted increasing attention due to their structural diversity, abundance, and low environmental impact, making them valuable for novel fungicide development. In this study, a series of coumarin-7-N/O acethydrazide derivatives were synthesized by incorporating acetic acid moieties into the coumarin scaffold. The in vitro antifungal activities these compounds evaluated against six common plant pathogenic fungi. Compound B13 demonstrated significant effects, with EC50 values 3.24, 1.89, 1.70, 0.36, 4.27, 1.50 μg/mL Botrytis cinerea, Alternaria solani, Fusarium graminearum, Rhizoctonia Colletotrichum orbiculare alternata, respectively. Notably, activity R. solani was comparable commercial boscalid (EC50 = 0.31 μg/mL), while A. alternata significantly higher than that 13.60 2.55 μg/mL, respectively), Furthermore, vivo assays protective efficacy at 200 (86.9%) Validamycin (92.1%). inhibitory effects further confirmed through scanning electron microscopy fluorescence microscopy. influence charge distribution compound on its investigated using Density Functional Theory (DFT). Abstract Structural modification is an effective way to improve the antifungal activity of natural products and has been widely used in development novel fungicides. In this work, a series aminocoumarin‐based Schiff bases were synthesized characterized by 1 H‐NMR, 13 C NMR HR‐MS spectra. The vitro inhibition all compounds was tested against four phytopathogenic fungi ( Alternaria solani , Fusarium oxysporum Botrytis cinerea alternata ) using mycelial growth rate method. results showed that most target exhibited significant activities. particular, 5b 5c 5d 5h 5n 7c 7n 7p more than commercially available fungicides, chlorothalonil azoxystrobin. structure‐activity relationship revealed electron‐withdrawing groups with electronegativity introduced at C‐3 position improving inhibitory halogenated benzaldehydes would be necessary preparation bases. compound (EC 50 =8.73 μg/mL) =26.25 much better positive controls (chlorothalonil azoxystrobin). Therefore, could serve as promising lead for broad‐spectrum which useful applications agriculture.
