Liquid chromatography coupled to high resolution mass spectrometry (LC-HRMS)-based methods have become the gold standard methodology for comprehensive profiling of human plasma lipidome. However, both complexity lipid chemistry and LC-HRMS-associated data pose challenges characterization this biological matrix. In accordance with current consensus quality requirements LC-HRMS lipidomics data, we aimed characterize NIST® Standard Reference Material Human Plasma (SRM 1950) using an LC-ESI(+/–)-MS method compatible high-throughput lipidome profiling. We generated a highly curated database increased coverage, quality, consistency, including additional assurance procedures involving adduct formation, within-method m/z evaluation, retention behavior species within chain isomers, expert-driven isomeric isobaric interferences. As proof-of-concept, showed utility our in-house LC-MS lipidomic –consisting 592 entries– fast, comprehensive, reliable from healthy volunteers. are confident that implementation robust resource will significant impact by reducing redundancy delays bottlenecks in untargeted studies.

The coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome 2 (SARS-CoV-2), has presented global challenges with a diverse clinical spectrum, including complications and systemic effects. This review explores the intricate relationship between mitochondrial dysfunction, aging, obesity in COVID-19. Mitochondria are vital for cellular energy provision resilience against age-related macromolecule damage accumulation. They manage allocation cells, activating adaptive responses stress signals such as redox imbalance innate immunity activation. As organisms age, function diminishes. Aging obesity, linked to compromise antiviral response, affecting release of interferons, worsening COVID-19 severity. Furthermore, development post-acute sequelae SARS-CoV-2 infection (PASC), also known long COVID been associated altered metabolism, chronic immune dysregulation derived from dysfunction. Understanding interplay mitochondria, viral infections provides insights into pathogenesis. Targeting health may offer potential therapeutic strategies mitigate outcomes address long-term consequences infected individuals.
