Abstract Rheumatoid arthritis (RA) is a debilitating autoimmune disease that causes chronic pain and serious complications, presenting significant challenge to treat. Promising approaches for treating RA involve signaling pathways modulation targeted therapy. To this end, multifunctional nanosystem, TPC‐U@HAT, has been designed therapy, featuring multitargeting, dual‐stimuli response, on‐demand drug release capabilities. TPC‐U@HAT composed of probe/prodrug TPC, JAK1 kinase inhibitor upadacitinib, the carrier HAT. TPC an aggregation‐induced emission (AIE)‐active NIR‐II chromophore TPY NF‐κB/NLRP3 caffeic acid phenethyl ester (CAPE), connected via boronic bond which serves as reactive‐oxygen‐species‐responsive linker. The carrier, HAT, created by grafting bone‐targeting alendronate hydrophobic tocopheryl succinate onto hyaluronic chains, can encapsulate upadacitinib form TPC‐U@HAT. Upon intravenous injection into mice, accumulates at inflamed lesions through both active passive targeting, overexpressed hyaluronidase H 2 O therein cleave polymer chains boronate bonds, respectively. This generates AIE‐active detection therapeutic evaluation optoacoustic imaging fluorescent concomitantly releases CAPE exert efficacious therapy inhibiting JAK‐STAT pathways.

Fluorescence analysis is an increasingly important contributor to the early diagnosis of kidney diseases. To achieve precise visualization kidneys and related diseases, asymmetric pyrrolopyrrolidone (DPP) dye platform with C-aromatic substituents N-lipophilic/hydrophilic modification was constructed. Based on these, we developed renal-clearable, water-soluble, injury biomarker leucine aminopeptidase (LAP) activated ratiometric fluorescent probe DPP-S-L. In mouse model cisplatin-induced acute during development type 2 diabetes diabetic disease, visualized for first time upregulation LAP in urine by dual-channel fluorescence signal diagnosed earlier more sensitively than blood/urine enzyme detection tissue analysis. Drug-induced renal failure (DIRF) poses a serious medical complication with high mortality risk. However, early diagnosis or prognosis of DIRF remain challenging, as current methods rely on detecting late-stage biomarkers. Herein we present library zwitterionic unimolecular hemicyanines (ZCs) available for constructing activatable reporters to detect since its initial stage. Zwitterionic properties these probes are achieved through interspersedly integrating alkyl sulfonates and quaternary ammonium cations onto hemicyanine skeleton, which result in record low plasma protein binding (<5 %) remarkable clearance efficiencies (≈96 %). An reporter ZCRR is further developed by masking the optimal candidate ZC6 tetrapeptide specifically cleavable caspase-8, an initiating indicator apoptosis. In living mice cisplatin-induced DIRF, systematically administered efficiently accumulates kidneys responds elevated caspase-8 near-infrared fluorescence signals 'turn-on', enabling sensitive detection intrarenal apoptosis 60 h earlier than clinical methods, precise evaluation remediation effects different medications mice.
