Copper oxide nanoparticles (CuO-NPs) are commonly used metal oxides. Betaine possesses antioxidant and neuroprotective activities. The current study aimed to investigate the neurotoxic effect of CuO-NPs on rats capability betaine mitigate neurotoxicity. Forty rats; 4 groups: group I a control, II intraperitoneally (0.5 mg/kg/day), III orally (250 mg/kg/day) CuO-NPs, IV for 28 days. Rats were subjected neurobehavioral assessments. Brain samples processed biochemical, molecular, histopathological, immunohistochemical analyses. Behavioral performance demonstrated increasing locomotion cognitive abilities. Group exhibited significantly elevated malondialdehyde (MDA), overexpression interleukin-1 beta (IL-1β), tumor necrosis factor-alpha (TNF-α). Significant decrease in glutathione (GSH), downregulation acetylcholine esterase (AChE), nuclear factor erythroid 2-like protein 2 (Nrf-2), superoxide dismutase (SOD). Histopathological alterations; neuronal degeneration, pericellular spaces, neuropillar vacuolation. Immunohistochemically, an intense immunoreactivity is observed against IL-1β glial fibrillary acidic (GFAP). partially neuroprotected associated alterations. A significant at MDA, IL-1β, TNF-α, increase GSH, upregulation AChE, Nrf-2, SOD. alterations ameliorated. Immunohistochemical intensity GFAP reduced.
