The treatment of ulcerative colitis (UC) has been a major medical challenge due to the lack safe and effective drugs. Molecular hybridization is promising strategy for development drugs with pleiotropic activity, which demonstrated in wide range diseases. Tofacitinib exhibited significant effects on remission UC, but series adverse have occurred during its clinical application. Herein, we propose utilize molecular link tofacitinib cytoprotective H2S donor (ADTOH) obtain hybridized molecules ZX-4C~ZX-6C. Among them, ZX-4C best performance release rate against dextran sulfate sodium (DSS)-induced injury. vivo studies showed that could effectively alleviate DSS-induced by enhancing oxidative stress defense reducing inflammatory response, demonstrating it more potent than parent data from present study support this provides avenue UC.
