Streptomyces sp. MJM3502 is a promising producer of rufomycins, which are class potent anti-tuberculosis lead compounds. Although the structure, activity, and mechanism main rufomycin 4/6 its analogs have been extensively studied, significant gap remains in our understanding genome sequence biosynthetic pathway MJM3502, metabolic engineering has not yet reported. This study established genetic manipulation platform for strain. Using CRISPR/Cas9-based technology to in-frame insert strong kasO∗p promoter upstream rufB rufS genes BGC, we increased production by 4.1-fold 2.8-fold, respectively. Furthermore, designing recombinant strains inserting encoding cytochrome P450 enzymes led new derivatives. These findings provide basis enhancing valuable natural compounds offer insights into generation novel active products via synthetic biology engineering.

Thalassemia is one of the inherited hemoglobin disorders worldwide, resulting in ineffective erythropoiesis, chronic hemolytic anemia, compensatory hemopoietic expansion, hypercoagulability, etc., and when a mother carries thalassemia gene, child more likely to have severe thalassemia. Furthermore, economic time costs genetic testing for prevent many patients from being diagnosed time. To solve this problem, we performed least absolute shrinkage selection operator (LASSO) regression analyze correlation between blood routine indicators containing mean corpuscular volume (MCV), (MCH), concentration (MCHC), red cell (RBC). We then built nomogram predict occurrence thalassemia, receiver operating characteristic (ROC) curve was used verify prediction efficiency model. In total, obtained 7,621 cases, including 847 6,774 non-thalassemia. Among patients, with positivity rate 67.2%, 569 cases were positive α-thalassemia, 31.5%, 267 β-thalassemia. The remaining 11 both α- Based on machine learning algorithm, screened four optimal indicators, namely, MCV, MCH, RBC, MCHC. AUC value MCHC 0.907, 0.906, 0.796, 0.795, respectively. Moreover, model 0.911. summary, novel effective which functioned accurately, may provide new insights early screening future.
