Granulocyte–macrophage colony-stimulating factor (GM-CSF) has many more functions than its original in vitro identification as an inducer of granulocyte and macrophage development from progenitor cells. Key features GM-CSF biology need to be defined better, such the responding producing cell types, links with other mediators, prosurvival versus activation/differentiation functions, when it is relevant pathology. Significant preclinical data have emerged deletion/depletion approaches indicating that a potential target inflammatory/autoimmune conditions. Clinical trials targeting or receptor shown encouraging efficacy safety profiles, particularly rheumatoid arthritis. This review provides update on above topics current issues/questions surrounding biology.

The cellular complexity of human brain development has been intensively investigated, although a regional characterization the entire cerebral cortex based on single-cell transcriptome analysis not reported. Here, we performed RNA-seq over 4,000 individual cells from 22 regions mid-gestation embryos. We identified 29 cell sub-clusters, which showed different proportions in each region and pons especially high percentage astrocytes. Embryonic neurons were as diverse adult neurons, they possessed important features their destinies adults. Neuron was unsynchronized cortex, dorsal appeared to be more mature than ventral at this stage. Region-specific genes comprehensively neuronal sub-cluster, large proportion these neural disease related. Our results present systematic landscape regionalized gene expression neuron maturation cortex.

While only recently developed, the ability to profile expression data in single cells (scRNA-Seq) has already led several important studies and findings. However, this technology also raised new computational challenges. These include questions about best methods for clustering scRNA-Seq data, how identify unique group of such experiments, determine state or function specific based on their profile. To address these issues we develop test a method neural networks (NN) analysis retrieval cell RNA-Seq data. We tested various NN architectures, some which incorporate prior biological knowledge, used obtain reduced dimension representation show that improves upon both, correctly experiments not training infer type by querying database tens thousands profiles. Such queries (which can be performed using our web server) will enable researchers better characterize when analyzing heterogeneous samples.
