Newborn neurons in the dentate gyrus of adult hippocampus rely upon cAMP response element binding protein (CREB) signaling for their differentiation into mature granule cells and integration network. Among its many targets, transcription factor CREB activates expression a gene locus that produces two microRNAs, miR-132 miR-212. In cultured cortical hippocampal neurons, functions downstream from to mediate activity-dependent dendritic growth spine formation variety pathways. To investigate whether and/or miR-212 contribute maturation dendrites newborn hippocampus, we inserted LoxP sites surrounding miR-212/132 specifically targeted deletion by stereotactically injecting retrovirus expressing Cre recombinase. Deletion caused dramatic decrease dendrite length, arborization, density. The may express up four distinct -212 reverse strands miR-132* -212*. Using ratiometric microRNA sensors, determined is predominantly active product neurons. We conclude required normal suggest this also participate other examples CREB-mediated signaling.

During the last two decades, microRNAs (miRNAs) emerged as critical regulators of gene expression. By modulating expression numerous target mRNAs mainly at post-transcriptional level, these small non-coding RNAs have been involved in most, if not all, biological processes well pathogenesis a number diseases. miR-132 and miR-212 are tandem miRNAs whose is necessary for proper development, maturation function neurons deregulation associated with several neurological disorders, such Alzheimer's disease tauopathies (neurodegenerative diseases resulting from pathological aggregation tau protein human brain). Although their involvement neuronal functions most described, evidences point towards role many other processes, including inflammation immune functions. Incidentally, was recently classified 'neurimmiR', class operating within between neural compartments. In this review, we propose an outline current knowledge about cells, by describing signalling pathways transcription factors regulating putative or demonstrated roles validated mRNA targets.
