Abstract The PI3K/AKT/mTOR (PAM) signaling pathway is a highly conserved signal transduction network in eukaryotic cells that promotes cell survival, growth, and cycle progression. Growth factor signalling to transcription factors the PAM axis regulated by multiple cross-interactions with several other pathways, dysregulation of can predispose cancer development. most frequently activated human often implicated resistance anticancer therapies. Dysfunction components this such as hyperactivity PI3K, loss function PTEN, gain-of-function AKT, are notorious drivers treatment disease progression cancer. In review we highlight major dysregulations cancer, discuss results AKT mTOR inhibitors monotherapy co-administation antineoplastic agents clinical trials strategy for overcoming resistance. Finally, mechanisms targeted therapies, including immunology immunotherapies also discussed.

CD69 is a membrane-bound, type II C-lectin receptor. It classical early marker of lymphocyte activation due to its rapid appearance on the surface plasma membrane after stimulation. expressed by several subsets tissue resident immune cells, including memory T (TRM) cells and gamma delta (γδ) therefore considered retention. Recent evidence has revealed that regulates some specific functions selected T-cell subsets, determining migration-retention ratio as well acquisition effector or regulatory phenotypes. Specifically, differentiation (Treg) secretion IFN-γ, IL-17, IL-22. The identification putative ligands, such Galectin-1 (Gal-1), suggests CD69-induced signaling can be regulated not only during cognate contacts between antigen-presenting in lymphoid organs, but also periphery, where cytokines other metabolites control final outcome response. Here, we will discuss new aspects molecular mediated involvement metabolic reprogramming regulating TH-effector lineages.

Metabolism drives function, on both an organismal and a cellular level. In T cell biology, metabolic remodeling is intrinsically linked to development, activation, differentiation, survival. After naive cells are activated, increased demands for currency in the form of ATP, as well biomass growth, proliferation, production effector molecules, met by rewiring metabolism. Consequently, pharmacological strategies being developed perturb or enhance selective processes that skewed immune-related pathologies. Here we review most recent advances describing changes occur during lifecycle. We discuss how metabolism can have profound effects health disease where it might be promising target treat variety

Abstract In recent years, immunotherapy represented by immune checkpoint inhibitors (ICIs) has led to unprecedented breakthroughs in cancer treatment. However, the fact that many tumors respond poorly or even not ICIs, partly caused absence of tumor-infiltrating lymphocytes (TILs), significantly limits application ICIs. Converting these “cold” into “hot” may ICIs is an unsolved question immunotherapy. Since it a general characteristic cancers resist apoptosis, induction non-apoptotic regulated cell death (RCD) emerging as new treatment strategy. Recently, several studies have revealed interaction between RCD and antitumor immunity. Specifically, autophagy, ferroptosis, pyroptosis, necroptosis exhibit synergistic responses while possibly exerting inhibitory effects on responses. Thus, targeted therapies (inducers inhibitors) against combination with exert potent activity, resistant This review summarizes multilevel relationship immunity RCD, including necroptosis, potential targeting improve efficacy malignancy.
