Over the past decade, Nomenclature Committee on Cell Death (NCCD) has formulated guidelines for definition and interpretation of cell death from morphological, biochemical, functional perspectives. Since field continues to expand novel mechanisms that orchestrate multiple pathways are unveiled, we propose an updated classification subroutines focusing mechanistic essential (as opposed correlative dispensable) aspects process. As provide molecularly oriented definitions terms including intrinsic apoptosis, extrinsic mitochondrial permeability transition (MPT)-driven necrosis, necroptosis, ferroptosis, pyroptosis, parthanatos, entotic death, NETotic lysosome-dependent autophagy-dependent immunogenic cellular senescence, mitotic catastrophe, discuss utility neologisms refer highly specialized instances these processes. The mission NCCD is a widely accepted nomenclature in support continued development field.

Detection of microbial DNA is an evolutionarily conserved mechanism that alerts the host immune system to mount a defense response infections. However, this detection also poses challenge as how distinguish foreign from abundant self-DNA. Cyclic guanosine monophosphate (GMP)-adenosine (AMP) synthase (cGAS) sensor triggers innate responses through production second messenger cyclic GMP-AMP (cGAMP), which binds and activates adaptor protein STING. cGAS can be activated by double-stranded irrespective sequence, including Although normally kept inactive in cells still not well understood, recent research has provided strong evidence genomic damage leads activation stimulate inflammatory responses. This review summarizes findings on instability trigger serves link inflammation, cellular senescence, cancer.

The recognition of DNA as an immune-stimulatory molecule is evolutionarily conserved mechanism to initiate rapid innate immune responses against microbial pathogens. cGAS-STING pathway was discovered important DNA-sensing machinery in immunity and viral defense. Recent advances have now expanded the roles cancer. Highly aggressive, unstable tumors evolved co-opt this program drive tumorigenic behaviors. In review, we discuss link between antitumor well cancer progression, genomic instability, tumor microenvironment, pharmacologic strategies for therapy. SIGNIFICANCE: defense infections. Given its role activating surveillance, it has been assumed that primarily functions a suppressor. Yet, mounting evidence suggests depending on context, signaling can also metastasis-promoting functions, chronic activation paradoxically induce immune-suppressive microenvironment.
