Unraveling the molecular mechanisms for COVID-19-associated encephalopathy and its immunopathology is crucial developing effective treatments. While an association between Parkinson's disease (PD) and viral infections has been recognized, the impact of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) on PD progression remains unclear. Here, we demonstrate that SARS-CoV-2 infection heightens risk using human embryonic stem cell (hESC)-derived dopaminergic (DA) neurons a angiotensin-converting enzyme (hACE2) transgenic (Tg) mouse model. Our findings reveal exacerbates susceptibility cellular toxicity in DA pre-treated with preformed fibrils (hPFFs). Additionally, nasally delivered infects hACE2 Tg mice, aggravating damage initiated by hPFFs. Mice infected display persisting neuroinflammation even after virus is no longer detectable brain. A comprehensive analysis suggests inflammatory response mediated astrocytes microglia could contribute to increased associated SARS-CoV-2. The recent SARS-CoV-2 pandemic has underscored the significance of viral infections, affecting billions lives and costing trillions dollars globally. Even beyond SARS-CoV-2, common infections with viruses like influenza, HIV, herpesviruses have profound impacts their typical manifestations, often triggering acute chronic pain syndromes that can be life-altering. These virally induced states arise through direct replication within neurons, or indirectly, via immune responses to infection in both contexts afferent signaling dorsal root ganglion (DRG) subsequent higher order integration intracranial systems.
