Crimean–Congo hemorrhagic fever (CCHF) is a serious tick-borne disease with wide geographical distribution. Classified as level 4 biosecurity risk pathogen, CCHF can be transmitted cross-species due to its aerosol infectivity and ability cause severe outbreaks high morbidity mortality. However, current methods for detecting anti-CCHFV antibodies are limited. This study aimed develop novel luciferase immunosorbent assay (LISA) the detection of CCHFV-specific IgG antibodies. We designed specific antigenic fragments nucleoprotein evaluated their sensitivity specificity in serum samples from mice horses. In addition, we compared efficacy our LISA commercial enzyme-linked (ELISA). Our results demonstrated that optimal antigen was located within stalk cut-off domain nucleoprotein. The exhibited indicated species significantly higher (at least 128 times) ELISA. proposed CCHFV-LISA has potential facilitate serological diagnosis epidemiological investigation CCHFV natural foci, providing valuable technical support surveillance early warning this disease.

The need for rapid and sensitive diagnostic tools is emphasized by the significant impact of infectious diseases on global health. This study presents a cell-free biosensor utilizing toehold switch technology, combined with nucleic acid sequence-based amplification (NASBA), high specificity sensitivity in Zika virus detection. switch, denovo-designed regulator gene expression, forms crux our detection system, offering versatile programmable approach to acid–based diagnostics. system based Escherichia coli extract served as platform sensor enabling real-time monitoring optimization reaction conditions minimal background leakage maximal activation efficiency. performance was rigorously evaluated through series tests, revealing that S23 demonstrated most promising effects sequence specificity. Notably, integration NASBA technology significantly enhanced sensitivity, achieving remarkable limit 2.9 aM, thus addressing intrinsic limitation switches detecting low-abundance targets. system's low cost, simplicity, adaptability various pathogens render it valuable asset health toolkit. advancement field synthetic biology, robust, sensitive, solution

Marburg virus disease (MVD) is a severe infectious characterized by fever and profound hemorrhage caused the (MARV), with mortality rate reaching 90%, posing significant threat to humans. MARV lies in its classification as biosafety level four (BSL-4) pathogen, which demands stringent experimental conditions substantial funding. Therefore, accessible practical animal models are urgently needed advance prophylactic therapeutic strategies for MARV. In this study, we constructed recombinant vesicular stomatitis (VSV) expressing glycoprotein (VSV-MARV/GP) utilized it surrogate induce lethal infection hamsters. Syrian hamsters infected VSV-MARV/GP presented symptoms such thrombocytopenia, lymphopenia, hemophilia, multiorgan failure, developing systemic akin that observed human patients, all animals succumbing at 2 3 days post-infection (dpi). Notably, pathogenicity of was found be species-specific, age-related, sex-associated, challenge route-dependent. Subsequently, efficacy MR191 monoclonal antibody validated model. summary, alternative model an effective tool rapidly screening medical countermeasures against GP vivo under BSL-2 conditions.
