Abstract Background: Hepatocellular carcinoma (HCC) presents challenges due to tumor heterogeneity and therapeutic resistance. Understanding the molecular mechanisms driving is crucial. Key transcription factors (HNF4A, HNF1A, FOXA1/2, etc.) signaling pathways (Wnt/β-catenin, FGF, HGF, are dysregulated in HCC. Dysregulation disrupts hepatocyte genetic programming, leading heterogeneous cell populations. Investigating these offers insights for targeted therapies improving patient outcomes Methods: Databases, including PubMed, MEDLINE, Google Scholar, open access/ subscription-based journals were searched published articles without any date restrictions, trace emergence of HCC by investigating hepatocyte-specific genes/TFs/signaling across cellular landscapes. Based on criteria mentioned methods section, studies systematically reviewed investigate Heterogeneity. This study adheres relevant PRISMA guidelines (Preferred Reporting Items Systematic Reviews Meta-Analyses). Results: into hepatocellular revealed dysregulation key (TFs) pathways. The tumor microenvironment (TME) plays an important role in the process of tumorigenesis, regulating growth, metabolism, proliferation, and invasion cancer cells, as well contributing to resistance conventional chemoradiotherapies. Several types cells with relatively stable phenotypes have been identified within TME, including cancer-associated fibroblasts (CAFs), tumor-associated macrophages (TAMs), neutrophils, natural killer (NK) which shown modulate cell metastasis, interaction immune system, thus promoting heterogeneity. Growing evidence suggests that tumor-cell-derived extracellular vesicles (EVs), via transfer various molecules (e.g., RNA, proteins, peptides, lipids), play a pivotal transformation normal TME into their protumorigenic counterparts. This review article focuses on functions EVs modulation view how exosomes contribute importance for diagnosis therapy.

Primary hepatocellular carcinoma (PHC) is the sixth most common cancer and third leading cause of death worldwide. Hepatocellular (HCC) accounts for 75%-85% PHC. LARP3 aberrantly expressed in multiple cancers. We found that it significantly highly liver tissues HCC patients, but exact role specific mechanism this abnormal expression are not yet clear. In study, through bioinformatics analysis, we concluded associated with a poor prognosis patients HCC. Through cellular experiments such as gene editing phenotypic functions, promotes occurrence development inhibits apoptosis. This investigation aimed to create a new nomogram based on complement C3 forecast 1-, 3-, and 5-year overall survival (OS) rates in patients with early-stage hepatocellular carcinoma (HCC) exhibiting microvascular invasion (MVI) post-curative surgery. study encompassed 1234 treated resection at the Affiliated Cancer Hospital of Guangxi Medical University. The cohort for primary included 865 from December 2015 2019, while validation comprised 369 patients. Follow-ups were conducted regularly until 2024. Variables predicting identified using Cox regression analyses, these, was constructed. nomogram's accuracy assessed via time-dependent ROC curves, calibration curves KM curve analyses. Investigations C3, PT, presence cirrhosis, tumor capsule, MVI-M2 as distinct predictors HCC Based these findings, predictive constructed validated, estimating OS. efficacy validated through analyses each demonstrating positive outcomes. Additionally, analysis effectively separated patient populations into two prognostic risk categories within both cohorts. The heterogeneity in prognostic survival and treatment response of hepatocellular carcinoma (HCC) limits the accurate assessment HCC-specific mortality. This study aimed to identify potential HCC subtypes through latent class analysis (LCA) improve mortality prediction optimize recommendations. We analyzed data from 7746 patients Surveillance, Epidemiology, End Results (SEER) databases, incorporating demographic clinicopathological information applying LCA subtypes. Prognostic across different were evaluated utilizing Cox proportional hazards regression competing risks models. classification was externally validated with 6791 patients. Four (LCAC1-LCAC4) determined. Compared LCAC1, both LCAC2 (HR = 1.887, p < .001) LCAC4 1.317, associated significantly shorter overall survival. had highest (HR: 2.395, .001), followed by 1.531, LCAC3 1.424, .001). lowest risk non-HCC-specific 0.613, Surgical treatment, particularly preoperative systemic therapy, improved all subtypes, whereas chemotherapy radiotherapy limited efficacy LCAC1 External validation corroborated these findings. provides a system that differentiates mortality, facilitating stratification recommendations, valuable insight for clinical decision-making.

In this study, we screened out the differentially expressed cuproptosis- ferroptosis-related genes (CFRGs) identified 17 informative prognosis-associated genes. A CFRG scoring model was constructed based on subtypes consensus clustering analysis principal component (PCA). Furthermore, immune profile, expression of checkpoint (ICGs) drug susceptibility were also compared between two score groups.
