Background: Hepatocellular carcinoma (HCC) is frequently diagnosed at late stages when curative treatments are no more appliable. Many studies have proved the active role of long non-coding RNAs (lncRNAs) in cancers’ biology; here, functional lncRNA NCK1-AS1 HCC was identified. Methods: Gene expression tumor tissues evaluated by examining online databases and 88 collected samples from our hospital. The interactions miR-22-3p with tyrosyl-tRNA synthetase (YARS) were tested conducting bioinformatics analysis, luciferase report, RNA pulldown experiments. CCK-8, colony formation, flow cytometry, wound healing, transwell experiments used to dissect NCK1-AS1/miR-22-3p/YARS axis HCC.Results: overexpressed cells tissues. Functional assays depicted that depletion hampered malignant character cells. controlled availability miR-22-3p, resulting YARS upregulation. found a clinical value for diagnosis. Moreover, rescue revealed inhibition or overexpression partially blocked function deficiency As downstream signaling pathway, we discovered activated PI3K/AKT miR-22-3p/YARS axis. Conclusion: present study verified could promote progression via activate signaling.

Increasing evidence suggests that taurine upregulated gene 1 (TUG1) is crucial for tumor progression; however, its role in hepatocellular carcinoma (HCC) and the underlying mechanisms are not well characterized. The expression levels of TUG1, miR-524-5p, sine oculis homeobox homolog (SIX1) were determined using quantitative real-time PCR. The regulatory relationships confirmed by dual-luciferase reporter assay. Cell proliferation invasion assessed Counting Kit 8 transwell assays. Glucose uptake, cellular lactate, lactate dehydrogenase (LDH), adenosine triphosphate (ATP) detected commercially available kits. Silencing TUG1 or SIX1 was performed lentivirus transduction. Protein measured immunoblotting.Cancer-associated fibroblasts (CAFs)-secreted exosomes promoted migration, invasion, glycolysis HepG2 cells releasing TUG1. promotive effects CAFs-secreted attenuated silencing targets miR-524-5p. knockdown inhibited miR-524-5p inhibitor. suppressed growth lung metastasis therefore increased survival xenograft model mice. We also found HCC patients with while decreased metastasis.CAFs-derived exosomal via miR-524-5p/SIX1 axis. These findings may help establish foundation development therapeutics strategies clinical management future.

As a crucial protumorigenic long noncoding RNA, colorectal tumor differential expression (CRNDE) has been confirmed to facilitate the progression of various cancers. However, its role in microenvironment (TME) hepatocellular carcinoma (HCC) is still unclear. Here we determined that CRNDE was upregulated HCC samples and CRNDE-positive cells were predominantly enriched malignant cells. In vivo functional assays revealed CRNDE-induced supported progression, recruited abundant granulocyte myeloid-derived suppressor (G-MDSCs) restricted infiltration T terms mechanisms, bound with Toll-like receptor 3 (TLR3) activated NF-κB signaling increase secretion c-x-c motif chemokine ligand (CXCL3).CRNDE knockdown could significantly suppress accumulation G-MDSCs enhance TME vivo.Taken together, our study reveals CRNDE-NF-κB-CXCL3 axis plays driving immunosuppressive niche by recruiting G-MDSCs.

Long non‑coding RNAs (lncRNAs) have recently gained attention due to their important roles in human cancer types, such as breast and gastric cancer. The present study measured alterations muskelin 1 antisense RNA (MKLN1‑AS) expression hepatocellular carcinoma (HCC) evaluated its clinical value patients with HCC. Additionally, the current investigated effects of MKLN1‑AS on malignant features HCC cells. detailed molecular mechanisms underlying cancer‑promoting activities cells were also elucidated. tissues cell lines was detected using reverse‑transcription quantitative PCR (RT‑qPCR). Cell Counting Kit‑8 assays flow cytometry used determine proliferation apoptosis. Migration invasion assays, well tumor xenograft experiments conducted analyze migration vitro growth vivo, respectively. interaction among microRNA‑654‑3p (miR‑654‑3p), hepatoma‑derived factor (HDGF) luciferase reporter assay, immunoprecipitation RT‑qPCR, western blotting rescue experiments. upregulated lines, a high associated shorter overall survival disease‑free Functionally, knockdown impaired proliferation, invasion, induced apoptosis vitro. Knockdown inhibited vivo. results indicated that functioned competing endogenous by sponging miR‑654‑3p targeting HDGF positively modulated MKLN1‑AS, partially abrogated this effect. Rescue demonstrated overexpression both abolished knockdown‑induced cellular processes In summary, pro‑oncogenic during progression serving sponge for increase expression.
