While immunotherapy with immune checkpoint inhibitors (ICIs) has revolutionized the clinical management of various malignancies, a large fraction patients are refractory to ICIs employed as standalone therapeutics, necessitating development combinatorial treatment strategies. Immunogenic cell death (ICD) inducers have attracted considerable interest partners for ICIs, at least in part owing their ability initiate tumor-targeting adaptive response. However, compared either approach alone, regimens involving ICD and not always shown superior activity. Here, we discuss accumulating evidence on therapeutic interactions between oncological settings, identify key factors that may explain discrepancies preclinical findings, propose strategies address existing challenges increase efficacy these combinations cancer.

An advanced photodynamic molecular beacon (PMB) was designed and synthesized, in which a distyryl boron dipyrromethene (DSBDP)-based photosensitizer Black Hole Quencher 3 moiety were connected via two peptide segments containing the sequences PLGVR GFLG, respectively, of cyclic peptide. These short are well-known substrates matrix metalloproteinase-2 (MMP-2) cathepsin B, both overexpressed wide range cancer cells either extracellularly (for MMP-2) or intracellularly B). Owing to efficient Förster resonance energy transfer between components, this PMB fully quenched native form. Only upon interaction with MMP-2 buffer solution cells, cleaved specifically, components could be completely separated, thereby restoring activities DSBDP moiety. This also activated tumors, it effectively suppressed tumor growth A549 tumor-bearing nude mice laser irradiation without causing notable side effects. In particular, did not cause skin photosensitivity, is very common effect therapy (PDT) using conventional "always-on" photosensitizers. The overall results showed that "double-locked" functioned as biological AND logic gate only unlocked by coexistence tumor-associated enzymes, greatly enhance specificity PDT.

Immunogenic cell death (ICD) refers to an immunologically distinct process of regulated that activates, rather than suppresses, innate and adaptive immune responses. Such responses culminate into T cell-driven immunity against antigens derived from dying cancer cells. The potency ICD is dependent on the immunogenicity cells as defined by antigenicity these their ability expose immunostimulatory molecules like damage-associated molecular patterns (DAMPs) cytokines type I interferons (IFNs). Moreover, it crucial host’s system can adequately detect adjuvanticity Over years, several well-known chemotherapies have been validated potent inducers, including (but not limited to) anthracyclines, paclitaxels, oxaliplatin. ICD-inducing chemotherapeutic drugs serve important combinatorial partners for anti-cancer immunotherapies highly immuno-resistant tumors. In this Trial Watch, we describe current trends in preclinical clinical integration chemotherapy existing immuno-oncological paradigms.
