COVID-19, caused by SARS-CoV-2, affects neuronal cells, causing several symptoms such as memory loss, anosmia and brain inflammation. Curcuminoids (Me08 e Me23) curcumin (CUR) are derived from Curcuma Longa extract (EXT). Many therapeutic actions have been linked to these compounds, including antiviral action. Given the severe implications of especially within central nervous system, our study aims shed light on potential curcuminoids against SARS-CoV-2 infection, particularly in cells. Here, we investigated effects CUR, EXT, Me08 Me23 human neuroblastoma SH-SY5Y. We observed that significantly decreased expression plasma membrane-associated transmembrane protease serine 2 (TMPRSS2) TMPRSS11D, consequently mitigating elevated ROS levels induced SARS-CoV-2. Furthermore, exhibited antioxidative properties increasing NRF2 gene restoring NQO1 activity following infection. Both effectively reduced replication SH-SY5Y cells overexpressing ACE2 (SH-ACE2). Additionally, all compounds demonstrated ability decrease proinflammatory cytokines IL-6, TNF-α, IL-17, while specifically INF-γ levels. Our findings suggest curcuminoid could serve a agent for impact context system involvement.

Abstract Although the COVID‐19 pandemic has officially ended, persistent challenge of long‐COVID or post‐acute COVID sequelae (PASC) continues to impact societies globally, highlighting urgent need for ongoing research into its mechanisms and therapeutic approaches. Our team recently developed a novel humanized ACE2 mouse model (hACE2ki) designed explicitly long‐COVID/PASC research. This exhibits human expression in tissue cell‐specific patterns akin Ace2. When we exposed young adult hACE2ki mice (6 weeks old) various SARS‐CoV‐2 lineages, including WA, Delta, Omicron, at dose 5 × 10 PFU/mouse via nasal instillation, demonstrated distinctive phenotypes characterized by differences viral load lung, trachea, turbinate, weight loss, changes pro‐inflammatory cytokines immune cell profiles bronchoalveolar lavage fluid. Notably, no mortality was observed this age group. Further, assess model's relevance studies, investigated tau protein pathologies, which are linked Alzheimer's disease, brains these post infection. findings revealed accumulation longitudinal propagation tau, confirming potential our preclinical studies long‐COVID.

ABSTRACT COVID‐19 exhibits not only respiratory symptoms but also neurological/psychiatric rarely including delirium/psychosis. Pathological studies on provide evidence that the cytokine storm, in particular (epidermal growth factor) EGF receptor (EGFR, ErbB1, Her1) activation, plays a central role progression of viral replication and lung fibrosis. Of note, SARS‐CoV‐2 virus (specifically, S1 spike domain) mimics directly transactivates EGFR, preceding inflammatory process. In agreement, anticancer drugs targeting EGFR such as Nimotuzumab tyrosine kinase inhibitors are markedly effective COVID‐19. However, these data might raise provisional caution regarding implication psychiatric disorder schizophrenia. The author's group has been investigating etiologic neuropathologic associations signaling with There significant molecular between schizophrenia ligand levels blood well brain. addition, perinatal challenges ligands intraventricular administration to rodents monkeys both resulted severe behavioral and/or electroencephalographic endophenotypes relevant this disorder. These animal models display postpubertal abnormality soliloquy‐like self‐vocalization intercortical functional connectivity. Long COVID, as currently defined by the World Health Organization (WHO) and other authorities, is a symptomatic condition that has been shown to affect an estimated 10 %-30 % of non-hospitalized patients after one infection. This damage, whether or not, lead various health impacts such heart attacks strokes. Outer membrane vesicles (OMVs) derived from Gram-negative bacteria naturally serve as a heterologous nano-engineering platform, functioning effective multi-use nanovesicles for diagnostics, vaccines, and treatments against pathogens. To apply refined OMVs human theranostic applications, we developed exposed receptor-binding domain (RBD) grafted with antigen 43 minimal modular system targeting angiotensin-converting enzyme 2 (ACE2). We constructed E. coli-derived using the autotransporter to display RBD referred viral mimetic Ag43β700_RBD OMVs. Based on this, protein were expressed onto Escherichia coli (E. coli) membrane.
