Bio-soft matter droplets formed via liquid-liquid phase separation (LLPS) of biopolymers have been found in living cells. Synthetic LLPS recently employed nanobiotechnology for artificial cell construction, molecular robotics, computing, diagnosis, and therapeutics. Controlling the dynamics bio-soft is essential developing such bio-inspired functional systems because maintain their functions based on temporally controlled biomolecular reactions assemblies. The mechanisms through which environmental conditions affect the expression of interconnected species is a key step to comprehending principles underlying complex chemical processes. In Nature, modifications triggered by environment have major impact on structure and function biomolecules regulate different reaction pathways. Abstract Proteinaceous inclusions formed by C9orf72 derived dipeptide-repeat (DPR) proteins are a histopathological hallmark in ~50% of familial amyotrophic lateral sclerosis/frontotemporal dementia (ALS/FTD) cases. However DPR aggregation/inclusion formation could not be efficiently recapitulated cell models for four out five DPRs. In this study, using optogenetics, we achieved chemical-free poly-PR condensation/aggregation cultured cells, with spatial and temporal control. Strikingly, nuclear condensates had anisotropic, hollow-centre appearance, resembling anisosomes aberrant TDP-43 species, their growth was limited RNA. These induced abnormal granulation the nucleus without activation stress response. Cytoplasmic aggregates that under prolonged light stimulation were more persistent than its condensates, selectively sequestered demixed state surrounded spontaneous granules. Our data suggest anisotropic condensation nucleus, causative dysfunction, may constitute an early pathological event C9-ALS/FTD. Anisosome-type represent common cellular pathology neurodegeneration previously thought. Highlights - Optogenetics can used to model cells.
