Cancer stem cells (CSCs) are a minority subset of cancer that can drive tumor initiation, promote progression, and induce drug resistance. CSCs difficult to eliminate by conventional therapies eventually mediate relapse metastasis. Moreover, recent studies have shown display plasticity renders them alter their phenotype function. Consequently, the varied phenotypes result in tumorigenesis, dissemination, drug-resistance potential, thereby adding complexity heterogeneity further challenging clinical management cancers. In years, microenvironment (TME) has become hotspot research owing its successful application immunotherapy. Notably, emerging evidence shows TME is involved regulating CSC plasticity. activate stemness pathways immune escape through cytokines exosomes secreted or stromal cells, inducing non-CSCs acquire properties increasing However, relationship between remains poorly understood. this review, we discuss investigations on illustrate underlying mechanisms potential implications suppressing progression We consider review help develop novel therapeutic strategies taking into account interlink

Significance Through the use of different in silico modeling approaches capturing tumor heterogeneity, we postulated that areas high metabolic activity would shift toward periphery as tumors become more aggressive. To confirm hypothesis and provide clinical value for finding, collected 18 F-FDG PET images breast cancers non–small-cell lung cancers, where measured distance from point maximum to centroid, normalizing it by a surrogate volume. The metric, NHOC, showed higher prognostic than other classical PET-based biomarkers used oncology, evidencing hotspot center its correlates with poor prognosis.

Studies at the molecular level demonstrate that dietary amino acid intake produces substantial effects on health and disease by modulating metabolism. However, how these may manifest in human food consumption patterns is unknown. Here, we develop a series of algorithms to map, characterize model landscape content food, patterns, individual including relations status, covering over 2,000 foods, ten 30,000 profiles. We find type acids contained foods highly dynamic with variability far exceeding fat carbohydrate. Some positively associate conditions such as obesity while others same negatively link disease. Using linear programming machine learning, show trade-offs can be accounted for satisfy biochemical constraints eating construct Pareto front practice, means achieving optimality face are commonly considered economic evolutionary theories. Thus this study enable design protein quality guidelines based quantitative framework.
