Aim: Our objective was to describe variations in attributable fractions (AFs) of hepatitis B virus (HBV) and C (HCV) primary liver cancer (PLC) across mainland China. Methods: We conducted a systematic review meta-analysis studies published up July 2024 PubMed, Embase, WanFang, China National Knowledge Infrastructure. Eligible reported the prevalence HBV HCV infection, alone combination, PLC. AFs HBV, HCV, non-viral etiology PLC were estimated by province and, when possible, sex, age, histological diagnosis, study periods. Regional overall weighting provincial regional population size. Publication bias heterogeneity assessed funnel plots cases, using Egger test Results: included 240 with 71,905 cases from 31 provinces, autonomous regions municipalities for 76.5%, 6.6%, 16.9%, respectively. The varied region, ranging 68.7% Northwest 82.9% South Central Respective ranged 12.7% Northeast down 3.7% Non-viral 13.4% 21.0% region. 57.9% intrahepatic cholangiocarcinoma (ICC), AF double ratio (PR), 2.11 (95%CI: 1.89-2.34) hepatocellular carcinoma (HCC, 76.3%) than ICC (37.3%) cases. Prevalence higher male (83.9%) female (74.6%) mean age at diagnosis 10 years (64.9 years) (54.4 years). Conclusion: substantially region China, as well sex which can inform strategies prevention control.

Lactate plays a critical role as an energy substrate, metabolite, and signaling molecule in hepatocellular carcinoma (HCC). Intracellular lactate-derived protein lysine lactylation (Kla) is identified contributor to the progression of HCC. Liver cancer stem cells (LCSCs) are believed be root cause phenotypic functional heterogeneity However, impact Kla on biological processes LCSCs remains poorly understood. Here enhanced glycolytic metabolism, lactate accumulation, elevated levels observed compared HCC cells. H3K56la was found closely associated with tumourigenesis stemness LCSCs. Notably, comprehensive examination lactylome proteome ALDOA K230/322 lactylation, which promoting Furthermore, this study demonstrated tight binding between aldolase A (ALDOA) dead box deconjugate enzyme 17 (DDX17), attenuated by ultimately enhancing regulatory function DDX17 maintaining This investigation highlights significance modulating its Targeting may offer promising therapeutic approach for treating

Background Dynamic changes in non-invasive tests, such as alanine aminotransferase (ALT) and MRI proton-density-fat-fraction (MRI-PDFF), may help to detect metabolic dysfunction-associated steatohepatitis (MASH) resolution, but a combination of tests be more accurate than either alone. We developed novel score, the MASH Resolution Index, histological resolution MASH. Methods This study included derivation cohort 95 well-characterised adult participants (67% female) with biopsy-confirmed who underwent contemporaneous laboratory testing, MRI-PDFF liver biopsy at two time points. The primary objective was develop score no worsening fibrosis. most predictive logistic regression model selected based on highest area under receiver operating curve (AUC), lowest Akaike information criterion Bayesian criterion. then externally validated distinct 163 from clinical trial. Results median (IQR) age body mass index were 55 (4562) years 32.0 (3037) kg/m 2 , respectively, cohort. (MASH Index) MRI-PDFF, ALT aspartate aminotransferase. had an AUC 0.81 (95% CI 0.69 0.93) for detecting calibrated well performed robustly external validation (AUC 0.83, 95% 0.76 0.91), outperformed MRI-PDFF. Conclusion Index useful non-invasively identify resolution.

Abstract Hepatocellular carcinoma (HCC) is one of the leading contributors to cancer-related mortality worldwide. Nop2/Sun domain family member 5 (NSUN5), a conserved RNA 5-methylcytosine methyltransferase, conventionally recognized as oncogenic. However, its role in HCC development remains unknown. In this study, we observed remarkable upregulation NSUN5 expression both tumor tissues from patients with HCC, establishing correlation unfavorable clinical outcomes. knockdown and overexpression significantly inhibited promoted cell proliferation, respectively. Additionally, employing combination methylated immunoprecipitation sequencing (MeRIP-seq) RIP-seq techniques, identified zinc finger BED domain-containing protein 3 (ZBED3) novel downstream target NSUN5. found that ZBED3 counteracted tumor-suppressing effect simultaneously reversed inhibition Wnt/β-catenin signaling pathway. summary, elucidated oncogenic ZBED3/Wnt/β-catenin pathway target. This study provides therapeutic for further treatment.

Heavy alcohol intake induces a wide spectrum of liver diseases ranging from steatosis, steatohepatitis, cirrhosis, and HCC. Although consumption is well-known risk factor for the development, morbidity, mortality HCC globally, alcohol-associated hepatocellular carcinoma (A-HCC) poorly characterized compared to viral hepatitis–associated Most A-HCCs develop after cirrhosis (AC), but direct carcinogenesis ethanol its metabolites A-HCC remains obscure. The differences between HCCs caused by other etiologies have not been well investigated in terms clinical prognosis, genetic or epigenetic landscape, molecular mechanisms, heterogeneity. Moreover, there huge gap basic research practice due lack preclinical models A-HCC. In current review, we discuss pathogenesis, heterogeneity, approaches, epigenetic, profiles A-HCC, insights into prospects future on potential effect cholangiocarcinoma metastasis also discussed.

The α-FAtE score, composed of alpha-fetoprotein, alkaline phosphatase, and eosinophil levels, has been reported as a predictor prognosis in hepatocellular carcinoma (HCC) patients treated with atezolizumab plus bevacizumab. This study aimed to investigate the predictive ability score for efficacy safety locoregional immunotherapy treatment HCC patients. We conducted retrospective 446 at Sun Yat-sen University Cancer Center from January 1st 2019 2023. performance was evaluated by concordance index, area under receiver operating characteristics curve, Kaplan-Meier curve multiple Cox regression analysis. were divided into 0-1 group (n=211) 2-3 (n=235). median progression-free survival(PFS) 7.3 months (95%CI 6.6-8.7 months), 12.3 (95% CI 10.4-14.1 months; P<0.001), respectively. overall survival (OS) 16.3 13.7-21.5 months) 34.1 27.6-NA had higher complete response (CR) rate experienced less drug-related adverse events than those group. Moreover, lower identified an independent prognostic indicator both OS PFS advanced receiving immunotherapy. is superior immunotherapy, offering valuable tool patient stratification planning.
